摘要
目的:通过分离纯化、培养及鉴定,获得人肝细胞肝癌组织中的肿瘤相关成纤维细胞(CAFs)。方法:采用组织块贴壁法获得原代细胞,通过差速酶消化法联合反复贴壁法进行纯化,再通过细胞形态观察和免疫细胞化学染色进行鉴定。结果:成功分离出纯度较高的CAFs。细胞多呈长梭形或星芒状,核圆形或卵圆形。免疫细胞化学染色示,细胞表达波形蛋白、α-平滑肌肌动蛋白和成纤维细胞活化蛋白。结论:成功分离纯化并鉴定CAFs,为下一步研究肝细胞癌相关成纤维细胞自身的生物学特性及其影响肝癌发生发展的机制提供了细胞学基础。
Objective:To obtain cancer-associated fibroblasts(CAFs) from human hepatocellular carcinoma byisolation,purification,culture and identification.Methods:Primary hepatocellular CAFs were obtained by tissueadhesion and purified by differential enzyme digestion and repeated adherent method. Then, the cells wereidentified using cytomorphological observation and immunocytochemical staining.Results:Highly purified CAFswere isolated successfully. Most CAFs were long-fusiform or star-shaped cells, with round or oval nuclei.Immunocytochemical staining revealed that the cells expressed vimentin,α-smooth muscle actin,and fibroblastactivation protein.Conclusions:The successful isolation, purification, and identification of CAFs build acytological basis for further study of the biological characteristics of hepatocellular CAFs and their impacts on thedevelopment and progression of hepatocellular carcinoma.
