摘要
目的研究东莞市2015—2016年手足口病(HFMD)病例中柯萨奇病毒A6型(CVA6)的流行情况及基因特征。方法采用RT-PCR方法对HFMD疑似病例标本进行检测,CVA6阳性标本上送广东省疾病预防控制中心,进行病毒分离及鉴定,扩增并测定VP1基因序列。从GenBank中选取其他CVA6代表株进行参考比对,利用DNASTAR软件进行核苷酸、氨基酸序列分析和同源性比较,并用Mega4.0.2软件构建亲缘性进化树。结果在1 145份标本中检出CVA6阳性标本580份,阳性率为50.66%(580/1 145)。序列比对显示,20株CVA6分离株VP1基因之间核苷酸同源性为93.6%~100.0%,氨基酸同源性为97.7%~100.0%。东莞市CVA6分离株VP1与近年来全球各地流行株核苷酸和氨基酸的同源性分别为93.4%~98.5%和97.0%~99.7%,与CVA6原型株Gdula以及我国山东省1996年分离株核苷酸和氨基酸相似性仅为82.5%~85.8%和94.1%~96.4%。系统进化树分析显示,2015—2016年东莞地区CVA6与近年来流行于世界各地的CVA6分离株同源性较高,而与原型株进化距离较远。结论 2015—2016年东莞市HFMD病原以CVA6为主,流行株为近年来流行于全国各地的CVA6新变种。
Objective To study the prevalence and genetic characteristics of coxsackievirus A6(CVA6) in DongguanCity from 2015 to 2016. Methods The specimens of hand foot and mouth disease(HFMD) cases were detected by the realtime RT-PCR method. The entire VP1 genes of CVA6 strains were amplified by RT-PCR and then sequenced, which werefinished by Guangdong CDC. The phylogenetic analysis between the strains isolated and those downloaded from Gene Bank wasdone by using the software DNASTAR and MEGA 4.0.2. Results Totally 580 out of 1 145 samples were CVA6 positive, andthe positive rate was 50.66%(580/1 145). The homogeneity of 20 CVA6 strains shared 93.6%-100.0% in nucleotide and97.7%-100.0% in amino acid identity. Compared with the new variant of CVA6 circulating in many places around the worldrecently, the nucleotide sequences had 93.4%-98.5% similarity and the amino acid sequence deduced had 97.0%-99.7%similarity. Compared with Gdula and the strains isolated from the epidemic in Shandong Province, China in 1996, thenucleotide and amino acid sequence similarity were 82.5%-85.8% and 94.1%-96.4% respectively. The analysis ofphylogenetic tree showed that CVA6 of Dongguan area from 2015 to 2016 had high homology with CVA6 isolates in recentyears, and was far from the evolution of the prototype plant. Conclusion From 2015 to 2016, the main aetiological agent ofcommon HFMD in Dongguan City is the new variant of CAV6.
