摘要
目的:探索无麻醉下豚鼠放射干预后鼻腔局部用药的动物模型的模式的建立。方法:100只豚鼠分为单纯对照组、麻醉后照射组、无麻醉照射组、麻醉后照射用药组、无麻醉照射用药组。单纯对照组不做任何干预,麻醉后照射组及麻醉后照射用药组用10%戊苯巴比妥腹腔注射用Co-60治疗机产生的射线作鼻部照射,照射剂量30Gy,每次300c Gy,每周5次,共照射10次;无麻醉照射组、无麻醉照射用药组在无麻醉下用固定器固定后照射给药。照射开始当天即开始用中药灌洗液对豚鼠鼻腔一天用药一次,共用药4周。采集照射后第1天、2周、4周、8周、12周颈总动脉处血液行肝肾功能检测,同时取鼻腔及鼻咽部黏膜组织常规光学显微镜下观察细胞形态,TUNEL法观察凋亡情况。结果:肝肾功能检测发现各组无显著性差异,光镜检查发现除单纯照射组外均有不同程度的炎症细胞,TUNEL法检查提示用药后凋亡细胞减少。结论:无麻醉下豚鼠局部固定能保持照射干预的稳定性,鼻腔局部用药能能稳定干预豚鼠鼻咽鼻腔组织的愈合状况,无麻醉下豚鼠固定鼻腔照射及药物干预是一个稳定的、可重复操作的动物模型。
Objective: To explore the Establishment of a guinea pig model of drug perfusion nasal after radiation on nasopharyngeal and nasal. Methods: 100 guinea pigs were divided into five groups randomly: The simple control group, the irradiation group after anesthesia, the irradiation group without anesthesia, the drug perfusion group after irradiation without anesthesia, the drug perfusion group after irradiation and anesthesia. Simple control group without any intervention. The anesthetics is 10% e phenobarbital. Irradiation method: Co-60 therapy equipment to produce a nose rays of light, exposure dose 30 gy, each 300 cgy, five times a week, a total of 10 times. Liver and kidney function test with the animal blood, at the same time in nasal cavity and nasal pharyngeal mucosa tissue of conventional optical microscope cell morphology, TUNEL method to observe apoptosis. Results: The liver and kidney function test found that each group there was no significant difference. The light microscope examination revealed except simple exposure group all have different degrees of inflammatory cells, TUNEL method revealed apoptosis cells reduce the medicine. Conclusion: Guinea pig local fixed without anesthesia can maintain the stability of the light interference, the local administration of nasal cavity can stable intervention in guinea pig nose pharynx nasal tissue healing condition, guinea pig without anesthesia fixed irradiation nasal cavity and drug intervention is a stable and repeatable operation of animal models.
