摘要
目的:对核桃楸皮黄酮的体内防治肝肿瘤活性进行研究,在初步明确其化学成分及药理药效的基础上,为其可能成为抗肝肿瘤候选药物的研发工作提供前期实验基础。方法:采用UPLC-Q-TOF-MS技术,结合质谱解析及对照品比对的方法,开展核桃楸皮黄酮化学成分初步指认研究,同时采用DEN致SD大鼠体内肝癌模型,开展核桃楸皮黄酮防治肝肿瘤药理药效研究。结果:通过相关方法,初步确认了核桃楸皮黄酮中的8种成分,推测了6种成分。同时药效学研究表明,核桃楸皮黄酮具有明确的体内防治肝肿瘤作用。在病理切片及甲胎蛋白病理指标上,核桃楸皮黄酮组与环磷酰胺组比较差异无统计学意义(P>0.05),但在死亡率、活动能力等生理指标上,核桃楸皮黄酮组明显优于环磷酰胺组。结论:核桃楸皮黄酮具有明确的体内防治肝肿瘤活性,与环磷酰胺组比较,有明确的死亡率低、毒副作用小等优势,为核桃楸皮黄酮可能成为抗肝肿瘤候选药物奠定了前期实验基础。
Objeetive: To Provide preliminary experimental basis to make Flavonoids from bark of Juglans mandshurica Maxim as the anti-liver tumor drug candidate through the study on the in vivo prevent liver tumor activity of it, based on the preliminary clarify of its chemical constituents and pharmacological activities. Methods: Carried on research about the preliminary identification of chemical constituents of Flavonoids from bark of Juglans mandshurica Maxim by using UPLC-Q-TOF-MS technology combined with method for mass spectrometry analysis and comparison of reference products, at the same time, the DEN induced liver cancer model in SD rats was used to carry on research about Pharmacological effect of prevent liver tumor from bark of Juglans mand-shurica Maxim. Results. Preliminary confirmation of the 8 components and infer of the 6 compon-ents inside Flavonoids from bark of Juglans mandshurica Maxim was carried out by some relevant methods. At the same time,pharmacodynamie studies showed that Flavonoids from bark of Juglans mandshurica Maxim had definite prevent liver tumor effect in vivo. Flavonoids from bark of Juglans rnandshu rica Maxim and Cyclophosphamide group have no definite difference ( P 〉 0. 05)in pathological section and pathological index of AFP. But when it comes to the physiological indexes of mortality and side effects, Flavonoids from bark of Juglans rnandshurica Maxim is better than Cyclophosphamide group. Conclusion : Flavonoids from bark of Juglans mandshurica Maxim has definite pre vent liver tumor activity in vivo, and has some advantages such as low mortality and small side effect compared with Cyclophospbamide group. This research provided preliminary experimental basis to make Flavonoids from bark of Juglans mandshurica Maxim as the antiliver tumor drug candidate.
