摘要
目的探讨清肝二十七味丸(寒水石、红花、石膏,等)对酒精性肝损伤(ALD)小鼠的保护作用。方法60只小鼠随机分为正常组,模型组,清肝二十七味丸高、中、低剂量组及联苯双酯组;模型组及各给药组小鼠均以白酒[10 m L/(kg·d)]、吡唑[25 m L/(kg·d)]、玉米油[2 m L/(kg·d)]混合物灌胃,早晚各1次;在2次灌胃间隔期间,灌胃给予清肝二十七味丸[1.6、0.8、0.4 g/(kg·d)]及联苯双酯[0.15 g/(kg·d)],造模周期为12周;检测谷丙转氨酶(ALT)、谷草转氨酶(AST)、羟脯氨酸(Hyp)、层粘连蛋白(LN)、透明质酸(HA)、丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、Wnt及β-连环蛋白(β-catenin)水平。结果与模型组比较,清肝二十七味丸高、中、低剂量组的血清ALT、AST、LN、HA水平及高、中剂量组的Hyp水平均显著性降低(P<0.05)。清肝二十七味丸组肝组织MDA水平显著降低,SOD及GSH-Px水平显著升高(P<0.05),各给药组肝组织Wnt及β-catenin蛋白的相对表达量均显著降低(P<0.05)。结论清肝二十七味丸对酒精性肝损伤小鼠具有保护作用,该作用与其具有的抗氧化及抑制Wnt/β-catenin信号通路激活作用有关。
AIM To explore the protective effects of Qinggan-27 Pills( Gypsum Rubrum, Carthami Flos,Gypsum Fibrosum,etc.) on mouse models of alcoholic liver damage( ALD). METHODS Sixty mice were randomly divided into normal group,model group,high,medium and low dose Qinggan-27 Pills groups and bifendate groups. The model and drug treatment groups were given the mixture of alcohol [10 m L/( kg · d) ],pyrazole[25 m L/( kg·d) ]and corn oil [2 m L/( kg·d) ]once every morning and evening. During the interval between two lavages,Qinggan-27 Pills [1. 6,0. 8,0. 4 g/( kg·d) ] and bifendate [0. 15 g/( kg·d) ] were lavaged for12 weeks and the levels of ALT,AST,Hyp,LN,HA,MDA,SOD,GSH-Px,Wnt and β-catenin were detected.RESULTS Compared with the model group,serum ALT,AST,LN and HA levels in high,medium and low dose groups and Hyp levels in high and middle dose Qinggan-27 Pills groups were significantly lower( P〈0. 05). Compared with the model group,Qinggan-27 Pills groups displayed significantly reduced MDA levels,significantly increased hepatic SOD and GSH-Px( P〈0. 05),and significantly reduced relative hepatic expression of Wnt and β-catenin as well( P〈0. 05). CONCLUSION Qinggan-27 Pills' protective effects on mouse models of ALD may be associated with its antioxidant behavior and inhibition of Wnt/β-catenin signaling pathway activation.
