摘要
建立了用超高效液相色谱-高分辨质谱法(UPLC-ESI-Q-TOF)测定糖果中碱性嫩黄O的分析方法。样品通过超声提取后,采用Oasis HLB固相萃取柱进行净化,洗脱液氮吹浓缩后定容至1 m L,直接用UPLC-ESI-Q-TOF检测。该方法的标准曲线在相关浓度范围(0.4~20.0)ng/m L内呈良好的线性关系,线性相关系数为R^2=0.9997,碱性嫩黄O的检出限为0.2μg/kg,加标回收率在80.85%~104.42%之间,样品重复测定8次,能得到较好的精密度,其RSD均小于10%。实验结果表明,该方法灵敏度高、重复性好、定量准确,适合于糖果中碱性嫩黄O的准确测定。
A method for determination of auramine O in candies using ultra performance liquid chromatography-high resolution mass spectrometry(UPLC-ESI-Q-TOF) for quantification was established. After ultrasonic extraction, the samples were purified by Oasis HLB solid phase extraction column, and the eluant was concentrated by nitrogen-blow, and make the sample volume to 1 m L, then detected by UPLC-ESI-Q-TOF. The standard curve of the method had a good linear relationship in the concentration range from 0.4 ng/m L to 20.0 ng/m L. The detection limit of the method was 0.2 μg/kg, the recovery was from 80.85%~104.42%. Candy samples were repeated 8 times with determination, which could get good precision, the RSD was less than 10%. The experimental results showed that, the method had high sensitivity, good repeatability and accuracy, fully suitable for determine the auramine O in candies.
