摘要
目的:采用网络药理学方法探索藏药多血康胶囊(DXK)治疗高原红细胞增多症(HAPC)的分子作用机制。方法:首先应用文献检索及TCMSP数据库分别筛选DXK中余甘子、红景天、沙棘、干姜4味单味药的化学成分,通过PharmMapper进行靶点的预测分析,将得到的潜在靶点导入MAS 3.0数据库进行靶点注释与分析,结合KEGG数据库进行通路分析,最后采用Cytoscape软件,构建DXK"化学成分-靶点"、"靶点-通路"网络图。结果:虚拟实验预测找到DXK含有的176个化学成分,涉及393个作用靶点,筛选后得到64个靶点和90条通路与DXK提高缺氧耐受力、抗氧化、改善血液流变学等作用相关。结论:DXK可能主要是通过调节免疫系统、炎症系统、心血管系统及代谢途径,并参与增强机体免疫、改善血液流变学及炎症反应、调节机体代谢等方面起到治疗HAPC的作用。
Objective: To explore the molecular mechanism of Tibetan medicine Duoxuekang capsule in the treatment of high altitude polycythemia(HAPC) by using network pharmacology.Methods: Firstly the chemical constituents of four flavor herbs(Phyllanthi Fructus,Rhodiolae Crenulatae Radix et Rhizoma,Hippophae Fructus,Zingiberis Rhizome) in Duoxuekang capsule were respectively screened by literature search and TCMSP database,then predictive analysis targets by Pharm Mapper server.Potential targets were submitted to MAS 3.0 database for target annotation and analysis.And pathway analysis would be performed with KEGG database.Finally,Cytoscape software was used to construct the"compounds-targets""targets-pathway"network diagram of Duoxuekang capsule in treatment of HAPC.Results: Virtual experiments predicted 176 chemical constituents of Duoxuekang capsule,which involved 393 action targets.Finally 64 targets and 90 pathways related to Duoxuekang capsule anti-hypoxia,anti-oxidation and blood stasis was got.Conclusion: Duoxuekang capsule in treatment of HAPC is mainly through the regulation of the immune system,inflammatory sydtem,cardiovascular system and metabolic pathways,and may be involved in enhancing immune,improve blood coagulation and inflammatory reaction,regulate metabolism and other aspects.
