摘要
目的:初步探讨蓝花参对硫酸钠致小鼠腹泻的抑制作用及其机制。方法:采用硫酸钠造成小鼠渗透性腹泻模型。观察小鼠生理状况及第1次排黑便时间;用碳粉推进实验及腹泻指数观察低、中、高剂量(2、6、18 g/kg)蓝花参在小鼠体内胃肠道的作用;HE染色法观察小肠病理变化;PCR观察结肠AQP4、NHE3 mRNA表达变化;采用离体动物实验法观察蓝花参对小鼠离体肠道平滑肌的作用。结果:与模型组比较,蓝花参高、中、低剂量均能改善腹泻小鼠成群蜷缩、嗜睡、毛发竖立情况,显著延缓第1次排黑便时间,降低小鼠腹泻指数及小肠推进率,改善小肠病理形态,上调腹泻小鼠结肠AQP4、NHE3 mRNA表达;并能增强小鼠离体平滑肌的振幅与张力。结论:蓝花参对硫酸钠致小鼠渗透性腹泻有抑制作用,该抑制作用与乙酰胆碱受体无关,其发挥抑制作用的机理可能为增加胃肠道对水、电解质的吸收。
Objective: To investigate the anti-diarrhea effect and the mechanism of Wahlenbergia marginata in mice with diarrhea induced by sodium sulfate.Methods: Diarrhea model was induced in mice by sodium sulfate.The physiological condition of mice and the time for their first melena were observed.The effect of Wahlenbergia marginata at different doses(2,6,18 g/kg) on gastrointestinal propulsion was evaluated by using the method of the charcoal meal and the observation of diarrhea indexes.HE staining was employed to observe the pathological changes of small intestine.PCR was used to detect the expression of AQP4,NHE3 and mRNA in colon.The effect of Wahlengergia marginata on intestinal smooth muscle of mice was also evaluated by in vitro experiment.Results: Compared with model group,Wahlenbergia marginata at different doses could significantly alleviate the diarrhea,reduce the symptom of huddling,sleepiness and bristling,and also postponed their first melena,and decreased the diarrhea indices and the rate of intestinal propulsion,ameliorated the pathological condition of small intestine,up-regulaled the mRNA expression of AQP4,NHE3 in colon and enhanced the amplitude and tension of isolated smooth muscle.Conclusion: Wahlenbergia marginata has an anti-diarrhea effect in mice with diarrhea induced by sodium sulfate,the inhibitory effect was not related to acetylcholine receptor,but the mechanism might be related to increasing gastrointestinal absorption of water and electrolytes.
