摘要
优化大球盖菇富硒发酵工艺,为饮食补硒提供新的有效途径。以大球盖菇菌丝生物量、有机硒和总硒浓度、菌丝体硒有机化程度和富硒率为考察指标,通过冷冻真空干燥、超微粉碎等技术进行前处理,采用单因素和正交试验方法,运用电感耦合等离子体质谱(Inductively coupled plasma mass spectrometry,ICP-MS)检测菌丝体有机硒和总硒浓度,优选大球盖菇富硒发酵工艺。结果表明:培养基为马铃薯200 g/L,葡萄糖20 g/L,酵母浸粉4 g/L,Na_2Se O_3·5H_2O 20 mg/L,KH_2PO_4 1.0 g/L,Mg SO_4 0.5 g/L。此时菌丝生物量为2.750 g/L,总硒浓度为339.168μg/g,有机化程度为86.68%。大球盖菇富硒培养时菌丝体硒有机化程度高,是可利用的新的补硒食用菌。
It was to investigate the optimization of selenium-enriched fermentation condition on Stropharia rugosoannulata. With the biomass, the content of selenium (Se) and organic Se, the rate of Se-accumulated and organic degree of mycelia as examining index, the contents of Se and organic were tested by ICPS-MS through vacuum freeze drying and superfine grinding technology of pretreatment. The liquid culture condition and liquid Se-enrichment condition were optimized in the single factor and orthogonal experiment. The results showed that the best liquid culture condition was as follows: potato 200 g/L, glucose 20 g/L, yeast extract powder 4 g/L, Na2SeO3·5H2O 20 mg/L, KH2PO4 1.0 g/L and MgSO4 0.5 g/L. The results indicated that the mycelia of Stropharia rugosoannulata could accumulate Se efficiently. On this condition, the contents of Se and organic degree in mycelia were 339.168 μg/g and 86.68%, and the yield of mycelia was 2.750 g/L. It indicated that Stropharia rugosoannulata was a suitable mushroom for organic Se-accumulated.
