摘要
以外消旋氟比洛芬和玉米淀粉为底物,固定化脂肪酶N435为催化剂,在异辛烷中对外消旋氟比洛芬进行一步酯化拆分,合成光学纯的(R)-氟比洛芬淀粉酯。通过单因素实验,研究反应温度、反应时间、氟比洛芬与淀粉质量比、酶添加量等对酶促酯化反应的影响。通过正交试验,确定制备氟比洛芬淀粉酯的最佳工艺条件:酶添加量5%、反应时间4 d、底物比1∶2、反应温度60℃,在此条件下,产物转化率为35%,对映体过量值EEp为55.45%。
Taking racemic flurbiprofen and corn starch as substrates and N435 immobilized lipase as catalyst,racemic flurbiprofen were split esterification in isooctane,step synthesis of optically pure( R)-flurbprofen starch.By a single factor experiment,this paper studied on the effect of reaction temperature,reaction time,flurbiprofen and starch mass ratio,the amount of enzyme on enzymatic deterification. On this basis,through the orthogonal test,the optimum conditions for preparing starch ester of flurbinprofen as follows: starch mass ratio of 5%,reaction time of 4 d,substrate ratio of 1 ∶ 2,temperature of 60 ℃,enzyme dosage under flurbiprofen and conversion of the product obtained was 35%,corresponding to the magnitude of the boby over EEp was 55. 45%.
