摘要
目的:观察黄连-吴茱萸组分配伍(小檗碱、吴茱萸碱配伍)对高脂血症模型大鼠肝脏Ob-Rb、JAK2、STAT3 mRNA表达的影响。方法:给予高脂饲料喂养建立高脂血症大鼠模型,筛选造模成功大鼠随机分为空白组,模型组,辛伐他汀组,黄连吴茱萸组分配伍高、中剂量组,分别给予相应药物灌胃治疗,干预8周后,检测各组动物血清TC、TG、HDL-C及LDL-C含量;检测各组动物肝指数及肝脏TC、TG水平;RT-PCR法检测肝脏Ob-Rb、JAK2、STAT3 mRNA表达。结果:黄连吴茱萸组分配伍中、高剂量组能显著降低高脂模型大鼠血清TC,LDL-C含量(P〈0.05或P〈0.01)、改善高脂模型大鼠肝脏脂肪病变,以及降低肝脏TG水平(P〈0.05)。与模型组相比,黄连吴茱萸组分配伍中剂量组肝脏Ob-Rb,JAK2,STAT3 mRNA表达明显下降(P〈0.05)。结论:黄连-吴茱萸组分配伍可能通过下调高脂模型大鼠肝脏Ob-Rb,JAK2和STAT3 mRNA的表达参与血脂调控。
Objective: To illustrate effects of Huanglian-Wuzhuyu component compatibility on expressions of hepatic leptin receptor( Ob-Rb),janus activated kinase signaltransducer 2( JAK2) and signal transducer and activator of transcription 3( STAT3) genes in hyperlipidemic rats. Method: After establishing the hyperlipidemic model successfully,SD rats were divided into normal group,model group,simvastatin group,Huanglian-Wuzhuyu component compatibility middle dosage group and Huanglian-Wuzhuyu component compatibility high dosage group. The model rats of hyperlipidemia were fed with high fat diet,while normal control rats were fed with fundamental diet. After 8 weeks treatment,serum triglycerides( TG),total cholesterol( TC),low-density lipoprotein cholesterol( LDL-C),high density lipoprotein cholesterol( HDL-C) and hepatic TC,TG and hepatic index were detected. RT-PCR method was used to detect the expressions of Ob-Rb,JAK2,and STAT3 genes in rats liver tissues. Result: The results showed that two different dosage of Huanglian-Wuzhuyu component compatibility groups significantly decreased TC and LDL-C contents in serum of the hyperlipidemic rats( P 0. 05),meanwhile reducing hepatic TG content of hyperlipidemic rats as well( P 0. 05),ameliorating steatosis in liver. Compared with the model group,Huanglian-Wuzhuyu component compatibility middle dosage group descended the expressions of Ob-Rb,JAK2 and STAT3 genes in hepatic tissues( P 0. 05). Conclusion: Huanglian-Wuzhuyu component compatibility could regulate the expressions of hepatic Ob-Rb,JAK2 and STAT3 genes in hyperlipidemic rats,which might be contributed to its effect of blood lipid levels of hyperlipidemia model rats.
