摘要
目的探讨糖尿病大鼠胃平滑肌细胞凋亡与哺乳动物雷帕霉素靶蛋(mTOR)及下游因子p70S6K、p-4E-BP1的变化及意义。方法制备糖尿病大鼠模型,分为正常对照(NC)组、模型2W(DM2W)组、模型4W(DM4W)组和模型6W(DM6W)组。流式细胞技术检测各组胃平滑肌细胞凋亡率,Western blot检测胃平滑肌p-mTOR、p-p70S6K、p-4E-BP1蛋白的表达。结果DM2W、DM 4W与DM 6W组胃平滑肌细胞细胞凋亡表现为逐渐增加,p-mTOR、p-p70S6K、p-4E-BP1表达均为先升高(与NC组比较,P<0.01)后降低(与DM4W组比较,P<0.01)。结论糖尿病大鼠胃平滑肌存在细胞凋亡与mTOR通路的变化,其中mTOR对细胞凋亡的调控不处于主导地位。
Objective To explore the role and significance of the apoptosis and the mTOR Signal pathway,including p70S6 Kand p-4 E-BP1,in gastric smooth muscle of diabetic rats. Methods SD rats were divided into normal control group(NC group),model two weeks group(DM2 W group),model four weeks group(DM4W group)and model six weeks group(DM6W group).The apoptosis rate of gastric smooth muscle in each group was detected by flow cytometry.The expression of p-mTOR,p-p70S6 Kand p-4 E-BP1 in each group was evaluated by Western blotting. Results The apoptosis rates gradually increased in DM2W,DM4W and DM6W groups.The protein expression levels of p-mTOR,p-p70S6 Kand p-4 E-BP1 firstly increased(compared with the normal group,P〈0.01)and then decreased(compared with 4W group,P〈0.01). Conclusion The apoptosis and mTOR pathway changes are exist in gastric smooth muscle in diabetic rats.The effect of mTOR pathway is not dominant in apoptosis regulation.
