摘要
香味基因是香稻育种的关键。本研究采用香味基因Xiang7F/7R对来源于辽宁省农科院和牡丹江市场的9个水稻品种进行序列分析。结果表明:(1)9个水稻品种在近400 bp处均出现一条带。(2)序列同源性分析发现,稼禾1号和龙粳香1号出现8 bp的片段缺失和3 bp的碱基突变。(3)日本晴与NCBI上发布的日本晴在该基因位点上存在5个碱基的插入/缺失,同源相似性达到99%。(4)遗传距离和系统发育树均表明,稼禾1号和龙粳香1号的遗传距离为0,聚为同一分支。以上结果表明,香味基因对不同水稻品种具有较高的灵敏度,可用于香稻品种的筛选,进而为后续的香稻品种选育奠定基础。
Fragrance gene is the key for fragrant rice breeding.In this research,a fragrance gene Xiang7F/7R was used to analyze the sequence of 9 rice varieties from the Liaoning Academy of Agricultural Sciences and Mudanjiang market.The results showed:(1) 9 rice varieties had only one band at about 400 bp.(2) Homology analysis of 9 kinds of rice sequences found that Jiahe 1 and Longgengxiang 1 had 8 bp deletions and 3 bp bases mutation.(3) There were five bases insertion/deletion of Nipponbare and Nipponbare published on NCBI at this genetic locus,and the homologous similarity reached 99 %.(4) Genetic distance and phylogenetic trees both indicated that the genetic distance between Jiahe 1 and Longgengxiang 1 was 0,which could be clustered into the same branch.The research results suggested that fragrance gene had high sensitivity to different rice varieties,which could be used for the selection of fragrant rice varieties,so as to lay a foundation for the further study of fragrant rice variety.
