摘要
本研究利用81对SSR引物和6对SRAP引物组合对23个中山杉品种及其父母本共26个基因型进行了遗传分析及指纹图谱构建。研究表明,至少利用7个SSR位点组合可有效区分22个基因型,所有81个SSR标记均不能准确区分中山杉149与302以及中山杉406与407,在此基础上,利用筛选出的2对SRAP标记可将未区分的4个无性系加以区分,最终构建了26个基因型的DNA指纹图谱,各基因型间遗传相似系数在0.40~0.96之间,通过聚类分析可将26个基因型分为两大类。研究结果为中山杉品种区分、鉴定及分子育种提供了重要的工具。
Genetic relationship and DNA-fingerprinting of 26 individuals of Taxodium (23 hybrids, 3 Male and Female Parents) were investigated using 81 SSR markers and 6 SRAP markers. The results showed that 22 clones could be effectively distinguished by 7 SSR loci combination, but the 4 clones remained could be only subdivided into two groups (T. ‘Zhongshanshan 149' and T. ‘zhongshanshan 302', T. ‘Zhongshanshan 406' and T. ‘Zhongshanshan 407') with 81 SSR. However, the 4 clones could be completely differentiated by two pairs of SRAP marker combination. Eventually, the DNA fingerprinting of all 26 clones was constructed with 7 SSR and 2 SRAP. The genetic similarities among individuals range from 0.40-0.96, on which, a phenetic tree showing the 26 clones were divided two clusters. The present results provided the valuable tools for the identification and molecular breeding of T. ‘Zhongshanshan'.
