摘要
目的观察黄精多糖对心脏重塑小鼠心脏组织中ICAM-1、VCAM-1蛋白表达的影响。方法皮下注射异丙肾上腺素,连续5天建立心脏重塑模型。灌胃给予黄精多糖治疗20天,实验结束后,取小鼠心脏检测心脏重量指数、观察心肌病理形态学变化,western blot测定心肌组织中ICAM-1、VCAM-1蛋白的表达。取血分离血清,生物化学法检测血清中超氧化物歧化酶(superoxide dismutase,SOD)、丙二醛(malondialdehyde,MDA)的含量;ELISA测定血清白细胞介素-6(interleukin6,IL-6)、肿瘤坏死因子-α(tumor necrosis factorα,TNF-α)的水平。结果与正常对照组相比,模型组心脏重量指数增加,血清中的IL-6、TNF-α含量增加,同时血清中MDA含量增加、SOD活性下降,心肌组织中ICAM-1、VCAM-1蛋白表达增加(P<0.01,P<0.05)。与模型组相比较,黄精多糖中、大剂量可显著降低心脏重量指数;降低血清中炎症因子IL-6、TNF-α的含量;增加血清中SOD活性,降低MDA含量;下调心肌组织中ICAM-1、VCAM-1蛋白表达(P<0.01,P<0.05)。结论黄精多糖可能通过抗氧化抗炎,降低心肌组织中ICAM-1、VCAM-1的蛋白表达保护异丙肾上腺素导致的小鼠心脏重塑。
Objective To observe the effect of polygonatum polysaccharides on the expression of ICAM-1 and VCAM-1 protein in cardiac remodeling mice. Methods Cardic remodeling mouse model was induced by subcutaneous injection with isoproternol for 5 consecutive days. Gavage of polygonatum polysaccharide for 20 days. After the experiment,the heart weight index of the mice was detected and the changes of myocardial pathomorphology were observed. The expression of ICAM-1 and VCAM-1 protein in myocardial tissue was measured by Western blot. Content of superoxide dismutase( SOD) and malondialdehyde( MDA) in the serum was measured by biological chemistry method; The level of interleukin-6( IL-6)and tumor necrosis factor-α( TNF-α) were measured by ELISA kit. Results Compared with control group,the heart weight index of model group was increased; TNF-α and IL-6 were increased,the activity of SOD was reduced,the content of MDA were increased,the expression of ICAM-1 and VCAM-1 in the myocardial tissue was increased. Compared with model group,Medium and high dose of polygonatum polysaccharides can significantly decrease the cardiac weight index,increase serum SOD activity and reduce serum MDA content,decrease the content of TNF-α and IL-6. The expressions of ICAM-1 and VCAM-1 were decreased. Conclusion Polygonatum polysaccharide may reduce the expression of ICAM-1 and VCAM-1 in myocardial tissue by antioxidation and anti-inflammatory to protect cardiac remodeling caused by isoprenaline.
