唐古特白刺叶化学成分及其抗炎活性研究 被引量:7

摘要 目的:对唐古特白刺Nitraria tangutorum Bor.叶的化学成分及其抗炎活性进行研究。方法:采用液液萃取、硅胶柱色谱和反相C18半制备液相色谱等方法进行分离纯化,根据理化性质及核磁共振波谱数据鉴定化合物的结构。采用脂多糖(LPS)诱导的小鼠巨噬细胞RAW264.7炎症模型,探究唐古特白刺叶中化合物5~10对巨噬细胞经LPS刺激后产生的炎症因子[肿瘤坏死因子-α(TNF-α)及白细胞介素-6(IL-6)]的影响,评价其抗炎活性。结果:从唐古特白刺叶的80%乙醇水提取物中分离得到10个化合物、分别鉴定为3,3',4'-三-O-甲基鞣花酸(1)、苜蓿素-7-O-β-D-葡萄糖苷(2)、邻苯二甲酸二丁酯(3)、鼠李秦素(4)、异杨梅二醇(5)、三春水柏枝素A(6)、(3S,5R,6S,7E)-3,5,6-三羟基-7-香波龙大柱烯-9-酮(7)、1,2,3,4-四氢-1,3,4-三氧代-β-咔啉(8)、3-羟基-β-咔啉(9)、β-咔啉-1-羧酸(10)。结论:化合物1~10均为首次从该属植物中分离得到。其中化合物5、6、8、9对炎症因子的抑制作用最强,其IC50值最小仅为19.16μmol·L-1,该研究首次发现唐古特白刺的抗炎成分。 Objective: To study the chemical components from the leaves of Nitraria tangutorum and its anti-inflammation bioactivity. Methods: The chemical components were isolated by liquid-liquid extraction,silica gel chromatography,and repeated C18 semi-preparative-HPLC. Their structures were elucidated by physicochemical properties and1 H-NMR and13 CNMR data. LPS induced RAW264. 7 inflammatory cell model was established to evaluate the effective inhibition of proinflammatory factor(TNF-α and IL-6) of compounds 5-10 from N. tangutorum. Results: Ten compounds were isolated from the ethanol extract of the leaves of N. tangutorum and their structures were identified as 3,3',4'-tri-O-methylellagic acid(1),tricin-7-O-β-D-glucopyranoside(2), dibutyl-phthalate(3), rhamnazin(4), Isomyricadiol(5), Myricarin A(6), trihydroxy-7-megastigmen-9-one(7),1,2,3,4-Tetrahydro-1,3,4-trioxo-B-carbo1 ine(8),3-hydroxy-β-carboline(9), β-carboline-1-carboxylic acid(10). Conclusion: All the isolates are reported from the plants of N. tangutorum. for the first time. Activity assay showed that compound 5,6,8 and 9 presented better inhibition for proinflammatory factor,the minimum IC50 is 19. 16 μmol·L-1.
