摘要
目的探讨PIWIL4在肝门部胆管癌(HCCA)中的m RNA及蛋白表达。方法收集经我院病理学检查确诊为HCCA的癌组织及癌旁(>1 cm)组织手术切除标本40例,采用聚合酶链反应(RT-PCR)及Western-blot印迹检测PIWIL4 m RNA和蛋白在癌组织及癌旁组织中的相对表达情况,并分析其与临床病理因素间的关系。结果 (1)40例样本中26例癌组织的PIWIL4表达明显高于癌旁组织,6例癌组织与癌旁组织差异不明显,8例癌组织中PIWIL4的表达低于癌旁组织。癌组织中PIWIL4的m RNA相对表达量高于癌旁组织(P<0.05)。(2)40例样本中26例患者肿瘤中的PIWIL4蛋白表达水平高于癌旁组织,14例患者肿瘤中的PIWIL4蛋白表达水平与癌旁组织差异不明显。(3)PIWIL4蛋白表达水平与肿瘤分化程度、肝内转移及门静脉癌栓有关(P<0.05)。结论 HCCA癌组织中PIWIL4基因处于激活状态,可将其作为一个诊治HCCA的新的靶点,且PIWIL4蛋白表达水平的增加可能与HCCA肿瘤恶性程度密切相关。
Objective To explore the PIWIL4 in the m RNA and protein expression in hilar cholangiocarcinoma( HCCA).Methods The resection specimens of HCCA cancer tissues and their para-carcinoma tissues( > 1 cm) confirmed by our hospital pathology were collected. And the m RNA and protein relative expression of PIWIL4 in cancer tissues and para-carcinoma tissues were detected by the polymerase chain reaction and Western-blot technique,and its relationship with clinicopathological factors was analyzed. Results I. Among the 40 cases of specimens,the PIWIL4 expression in 26 cases of cancer tissues was significantly higher while the PIWIL4 expression of 8 cases was lower than that in the para-carcinoma tissues,and there was no difference in the 6 cases of cancer tissues and their para-carcinoma tissues. The average m RNA relative expression of PIWIL4 in cancer tissues was significantly higher than that in the para-carcinoma tissues( P < 0. 05). II. The protein expression of PIWIL4 in 26 cases of cancer tissues was significantly higher than that in para-carcinoma tissues,and there was no significant difference in the protein expression of PIWIL4 between the 14 cancer tissues and their para-carcinoma tissues. III. The protein expression of PIWIL4 was correlated with tumor differentiation,intrahepatic metastasis and portal vein cancer embolus( P < 0. 05). Conclusion The PIWIL4 gene in HCCA cancer tissues is in the active status and can be used as a new target for the diagnosis and treatment of HCCA,and the increase of PIWIL4 protein expression may be closely related to the malignancy degree of HCCA tumor.
