摘要
试验选用黄粱木叶片,烘干后以一定的料液比在不同温度、功率下超声波处理一定时间,提取叶片中蛋白质后测定提取液中蛋白含量,运用正交分析法优化超声波提取黄粱木叶蛋白工艺。结果表明,料液比、超声波功率、处理温度、处理时间对叶蛋白提取率影响均极显著(p<0.01),各因素对提取率影响大小为料液比>温度>时间>功率,最优提取工艺为料液比1︰160(g/m L)、温度75℃、时间1.5 h、功率100 W,提取率达10.15%。该工艺能高效提取黄粱木叶片中蛋白质,为实际生产提供一定理论依据。
The aim of this study was to investigate the optimum ultrasonic-assisted process for the extraction of leaf protein from leaves of Anthocephalus chinensis. The leaves which were collected from South China Agricultural University were dried in an oven. The leaf protein was extracted at different temperature, different material ratio, different ultrasonic power and different extraction time. And the leaf extract protein content was also determined. The orthogonal analysis was used to optimize the ultrasonic extraction of the leaf protein. The results showed that the effect of temperature, material ratio, ultrasonic power and extraction time on leaf protein extraction efficiency were all extremely significant (p〈0.01). The effect of each factor on the extraction efficiency is the material ratio 〉 temperature 〉 time 〉power. The optimal extraction process was material ratio of 1 : 160 (g/mL), temperature of 75 ℃, time of 1.5 h, power of 100 W. The extraction efficiency was 10.15%. The process could efficiently extract the protein from the leaves ofAnthocephalus chinensis and provide a theoretical basis for the actual production.
