环孢菌素A治疗高血糖大鼠脊髓损伤的疗效探究 被引量:1

Efficacy of cyclosporin A in the treatment of spinal cord injury in hyperglycemic rats

摘要 目的探讨环孢菌素A(eyclostmrinA,CsA)对高血糖大鼠脊髓损伤(spinalcordinjury,SCI)的治疗效果。方法40只雌性SPF级SD大鼠随机分为4组:似手术组、SCI组、糖尿病SCI组和糖尿病SCI+CsA组,每组10只。监测各组大鼠造模前后血糖和体质量的变化,采用斜板实验法和BBB评分法检测各组大鼠运动功能的变化,同时分别采用免疫组织化学染色(损伤相关指标Calpain1)、流式细胞术、ELISA(炎性反应相关因子IL-10和TNF-α)、Westernblotting{线粒体损伤标记分子CYP-D和AIn检测各组大鼠损伤程度及CsA对高血糖合并SCI大鼠的治疗效果。结果同假手术组相比,SCI组、糖尿病SCI组calpain1、IL-10、TNF-α、CYP—D、AIF表达显著上升,细胞凋亡比例显著增多;糖尿病SCI组同SCI组相比,Calpain1、IL-10、TNF-α、CYP-D、AIF表达水平显著上调,细胞凋亡比例显著增多;但糖尿病SCI+CsA组同糖尿病SCI组相比,Calpaln1、IL-10、TNF-α、CYP-D、AIF表达水平显著下降,细胞凋产比例显著降低。结论高血糖可以明显加重SCI的继发性损伤,CsA可有效改善大鼠的运动能力,减少SCI后脊髓炎性反应、细胞凋亡及线粒体损伤,CsA能够有效缓解高血糖加重的SCI,可为高血糖的SCI患者提供一种新的和相对有效的治疗方法。 Objective To explore the curative effects of cyclosporin A (CsA) in the treatment of spinal cord injury (SCI) in hyperglycemic rats. Methods Forty female SPF SD rats were randomly divided into four groups: sham group, SCI group, hyperglycemic SCI group and hyperglycemic SCI+CsA group with 10 rats in each group. The changes of blood glucose and body weight of the rats in each group were monitored before and after modeling. The changes of rats' movement function were detected by inclined plate test and BBB score. Simultaneously, immunohistochemical staining (injury related indicators Calpain 1), flow cytometry, ELISA (inflammatory response related factors IL-10 and TNF-α) and Western blotting (mitochondrial injury biomarkers CYP-D and AIF) were used to detect damage degree of the rats in each group and the treatment effect of CsA on hyperglycemic SCI rats, respectively. Results Compared with sham group, the expression levels of Calpain 1, IL-10, TNF-α, CYP-D and AIF were significantly increased in SCI group and hyperglycemic SCI group, as well as the apoptosis ratio. Compared with SCI group, hyperglycemic SCI group showed higher levels of Calpain 1, IL-10, TNF-α, CYP-D, AIF and cell apoptosis rate. However, compared with hyperglycemic SCI group, the expression levels of Calpain 1, IL- 10, TNF-α, CYP-D and AIF decreased significantly, as well as the cell apoptosis rate, in the hyperglycemic SCI+CsA group. Conclusion Hyperglycemia can significantly increase secondary injury of SCI, while CsA can effectively improve the rats' movement function, reduce the inflammation responses of spinal cord after SCI, apoptosis and mitochondria damage. Therefore, CsA can effectively alleviate SCI aggravated by hyperglycemia, which can provide a new and potent therapeutic method for hyperglycemic SCI patients.
