摘要
SIRT1是Sirtuin基因家族成员之一,是一种具有烟酰胺腺嘌呤二核苷酸(NAD+)依赖性脱乙酰酶活性的组蛋白去乙酰化酶。帕金森病(PD)属于运动障碍性疾病,α-突触核蛋白的错误折叠、聚集及多巴胺能神经元的缺失是该病的重要病理改变。大量研究证实SIRT1通过去乙酰化组蛋白及众多转录因子,如p53、PGC-1α等,抵抗α-突触核蛋白的神经毒性,对PD产生神经保护作用。
SIRT1, a member of the Sirtuin family, is a histone deacetylase with nieotinarnide adenine dinucleo- tide (NAD+) dependent deacetylase activity. Parkinson's disease is a type of movement disorders. The misfolding, aggre- gation of α-synuelein and the loss of dopaminergic neurons are important pathological changes in PD. A large number of studies confirmed that SIRT 1 display neuroprotective effects against α-synuclein neurotoxicity through deacetylating his- tone and numerous transcription factors including p53, PGC-1α, and others.
