摘要
目的:探讨旋覆代赭汤对功能性消化不良(FD)大鼠的药效作用及其机制。方法:40只雄性SD大鼠按体质量随机分为正常组、模型组、阳性药组(多潘立酮)、旋覆代赭汤低剂量组、旋覆代赭汤高剂量组,采用"不规则喂养法配合夹尾刺激法"建立FD模型,连续给药14 d,每周称量体质量。分别在造模结束、给药结束时代谢笼法测定各组大鼠24 h代谢值。末次给药后,用半固体营养糊法检测胃排空率;大鼠麻醉后心脏取血,硝酸还原酶法检测血清一氧化氮(NO)含量、酶联免疫法检测血浆胃泌素(GAS)、胃动素(MTL)、生长抑素(SS)、血管活性肠肽(VIP)水平。取胃窦,HE染色后光镜下观察胃窦组织病理变化;免疫组织化学法检测胃窦组织中C-kit及TFF1表达。结果:与正常对照组比较,模型组大鼠体质量、24 h尿量、血浆GAS及MTL含量、胃排空率均显著降低(P<0.05),胃窦C-kit及TFF1的表达均显著减弱,24 h摄食量、饮水量、血清NO、血浆SS及VIP含量均显著升高(P<0.05);与模型组比较,各给药组24 h摄食量、饮水量、血清NO、血浆SS、VIP含量均显著降低(P<0.05),24 h尿量、血浆GAS及MTL含量、胃排空率均显著升高(P<0.05),胃黏膜C-kit、TFF1的表达均极为显著升高。结论:旋覆代赭汤可以明显地提高FD大鼠血浆GAS及MTL含量、促进胃排空及增强胃黏膜C-kit及TFF1的表达,降低血浆SS、VIP含量及血清NO含量可能是其治疗FD的作用机制。
Objective: To explore the pharmacodynamic action of Xuanfu Daizhe decoction on functional dyspepsia (FD) rats and mechanism. Methods :40 SD rats were randomly divided into five groups: model control group, high dose of Xuanfu Daizhe Tang group, low dose of Xuanfu Daizhe Tang group, domperidone group and normal control group, FD models were induced by moderately enraging the rats' tail and irregular feeding. Drugs were administrated for 14 days, the rats were weighed weekly. The 24 h metabolic rate was measured by metabolic cage on 15th day and 28th day. Gastric emptying was detected with phenolsul - fonphthalein at the end of treatment ; rats were executed after anesthesia, Gastrin ( GAS ), Motilin ( MTL), Somatostatin ( SS), Vasoactive intestinal peptide(VIP) and Nitric Oxide (NO)were detected by Enzyme Linked Immunosorbent Assay and Nitrate reduction method with the blood drawn from heart. The gastric antrum was observed under light microscope for pathological change after HE staining. The expression of C - kit and TFF1 in gastric antrum were detected by immune histochemical method. Results: Compared with the normal control group, body weight, 24 h total urine volume, gastric emptying were reduced( P 〈 0. 05 )in the model group, the expressions of C -kit and TFF1, the levels of plasma GAS and MTL were lowered, while, the 24 h food and water intake were increased( P 〈 0. 05 ), the levels of serum NO, plasma SS and VIP were raised. Compared with the model group, the 24 h food and water intake were decreased( P 〈0. 05 )in the Xuanfu Daizhe Tang groups and domperidone group, the levels of serum NO and plasma SS and VIP were lowered; however, 24 h total urine volume, gastric emptying were increased (P 〈 0.05 ), the expressions of C - kit and TFF1, the levels of plasma GAS and MTL were increased. Conclusion: Xumffu Daizhe Tang can promote the gastric emptying in FD rats, and its mechanism may be up -regulation the expressions of C - kit and TFF1 and the levels of plasma GAS and MTL, down - regulate the levels of serum NO and plasma SS and VIP.
