肾炎消白颗粒对糖尿病肾病大鼠足细胞podocin、nephrin表达的影响 被引量:3

Effects of Shenyan Xiaobai Granules( 肾炎消白颗粒) on Nephrin and Podocin Protein Expressions in Podocytes of Diabetic Nephropathy Rats

摘要 目的:通过实验观察肾炎消白颗粒对糖尿病肾病(DN)模型大鼠足细胞podocin、nephrin表达的影响,探讨其作用机制。方法:给予高脂、高糖饮食联合单侧肾脏切除后一次性腹腔注射链脲佐菌素(STZ)的方法建立DN大鼠模型。将大鼠随机分为正常对照组、模型对照组、盐酸贝那普利(洛汀新)组、肾炎消白颗粒高剂量组、肾炎消白颗粒低剂量组,共5组。连续灌胃给药8周,检测大鼠尿蛋白排泄率(UAE)、血清总胆固醇、三酰甘油;Western-Blot检测大鼠足细胞podocin和nephrin的表达。结果:肾炎消白颗粒能明显降低DN大鼠血清总胆固醇、三酰甘油水平及UAE,与模型组比较有显著性差异(P<0.05),低剂量组作用更显著,与西药对照组比较差异显著(P<0.05);与正常组相比,模型组大鼠肾组织podocin、nephrin表达水平显著降低(P<0.05);与模型对照组比较,肾炎消白颗粒组大鼠肾组织podocin、nephrin表达水平显著升高(P<0.05)。结论:肾炎消白颗粒可减少DN大鼠尿蛋白的排泄率,降低血脂水平,上调podocin和nephrin蛋白是其延缓DN进展的作用机制之意。 Objective: To observe the effects of Shenyan Xiaobai Granules on the expressions of podocin and nephrin protein in podocytes of diabetic nephropathy (DN) rats, and investigate the mechanism of Shenyan Xiaobai Granules in the prevention and treatment of DN. Methods: DN models were built by high fat/high sugar diet in combination with unilateral nephrectomy, giving intraperitioneal injection of streptozotocin for one time. Rats were divided randomly into 5 groups, blank control group, model control group, high dose Shenyan Xiaobai Granules group, low dose Shenyan Xiaobai Granules group, the group with Lotensin group, rats of each group was given the corresponding medicine continuously for 8 weeks, UAE, and the levels of total cholesterol (TC) and triglyceride(TG) during the $ weeks were determined, podocin and nephrin protein expressions in podocytes were detected by Western Blot. Results: UAE and serum TC, TG levels in Shenyan Xiaobai Granules groups were decreased, the effects of low dose Granules group were better that was more significance that those of West- eru medicine control group (P 〈 0. 05 ). The podocin and nephrin protein expressions in podocytes of model group were lowered to compared with normal group, those in Shenyan Xiaobai Granules groups were raised, the differences between model and Granules groups were significance ( P 〈 0. 05 ). Conclusion : Shenyan Xiaobai Granules can decrease UAE of DN rats,lower serum lipid, up - regulating podocin and nephrin protein expressions in podocytes was one of the mechanism that delaying progression of DN.
