摘要
目的了解矽尘对作业工人叉头转录因子3(forkhead/winged helix transcription factor,FOXP3)基因启动子区域DNA甲基化状态的影响。方法选取健康检查者和矽尘作业工人为对照组和接尘组,采用亚硫酸氢盐测序法检测外周血FOXP3基因启动子特定区域DNA甲基化水平。结果在男性中,无论对照组还是接尘组FOXP3基因启动子区(+2071,+2182)5个Cp G位点均呈现高甲基化(100%)。在女性中,对照组第1个Cp G位点(+2102)和第4个Cp G位点(+2144)呈现低甲基化(40%),而接尘组相关Cp G位点可见高甲基化(100%)。结论女性接尘作业工人FOXP3基因启动子特定位点DNA高甲基化可能与矽尘的毒性有关。
[Objective]To investigate the effect of silica dust on DNA methylation in the promoter of transcription factor forkhead box protein 3(FOXP3) gene in the exposed workers.[Methods] Then the level of DNA methylation in the particular region of the FOXP3 gene promoter was in the peripheral blood detected by bisulfite sequencing.[Results] In males,hypermethylation(100%)was observed in 5 Cp G sites of FOXP3 gene promoter(+2071,+2182) in the control group as well as in the exposure group.In females,the first Cp G site(+2102)and the fourth Cp G site(+2144)were found to be hypomethylated(40%) in the control group,while those in the exposure group were hypermethylated(100%).[Conclusion] The hypermethylation in the particular sites of the FOXP3 gene promoter might be relevant with the toxicity induced by silica dust in the female workers.
