摘要
目的 从小鼠树突状细胞(DC2.4)中提取外排体并载入盐酸阿霉素,研究外排体的细胞摄取能力与肿瘤细胞杀伤效率。方法 使用超速离心的方法从DC2.4细胞的培养基上清液中提取外排体,并通过透射电子显微镜验证提取结果;通过电穿孔法载入盐酸阿霉素并测定其粒径和Zeta电位;用超速离心法进行纯化,测定其载药量及包封率;选用小鼠乳腺癌细胞(4T1)为细胞模型,考察制剂的摄取能力和杀伤力。结果 成功制备了以内源性物质外排体为载体的给药系统,相比于传统给药系统脂质体,在4T1细胞中的入胞能力以及杀伤力都有显著性增加。结论 以外排体为载体的新型给药系统在未来的抗肿瘤治疗领域具有巨大的潜力。
OBJECTIVE To extract exosomes from DC2.4 cell line, and then to load Doxorubiein into exosomes and to investigate the cellular uptake and tumor cell killing effect. METHODS Using ultraeentfifugation method to extract exosomes from the culture supernatant of DC2.4 cells and validating the results by a transmission electron microscope,loading Doxoruhicin by electroporation and determining the particle size and Zeta potential. Then purified by ultracentrifugation, determining drug loading and encapsulation efficiency. Mice mammary cells (4T1) were used to study the cellular uptake and tumor cell killing effects. RESULTS The exosomes drug delivery system was successfully prepared, comparing to liposomes, the cellular uptake and tumor cell killing effects were increased significantly in 4T1 cells. CONCLUSION Exosomes drug delivery system has great potential in the future in anti - tumor therapeutics.
