摘要
目的:探讨淫羊藿苷对气管切开插管大鼠肺部肥大细胞活性及炎性反应的影响。方法:将SPF级健康雄性SD大鼠分成正常对照(A)组、气管切开插管留置模型非用药(B)组、气管切开插管淫羊藿苷用药(C)组。造模成功6h后C组予淫羊藿苷灌胃,A、B组予等量0.9%氯化钠溶液。于24、72、168h 3个时间段每组分别处死6只大鼠进行取材检测。结果:光镜下,B、C组肺组织病理各个时段均有不同程度的炎性反应等改变,Smith评分均高于A组(P<0.05);但C组病变明显小于B组(72h后,P<0.05)。透射电镜下B组各时段内肥大细胞不同程度出现脱颗粒,24h时最明显,但C组程度明显低于B组(P<0.05)。B、C组β-类胰蛋白酶表达在3个时段均高于A组(P<0.05);从72h后C组明显低于B组(P<0.05),以168h为最明显。B、C组组胺含量在3个时间段高于A组(P<0.05),B、C两组比较无差异。B、C两组TNF-α、IL-6含量变化基本一致,在不同时间段均明显高于A组(P<0.05),24h时达到峰值,随后下降。结论:淫羊藿苷可减轻气管切开插管大鼠肺部炎性反应,其机制与抑制肥大细胞活性有关。
Objective: To study the effects of pulmonary mast cell's activation and inflammation in rats model with tracheotomy intubation by one-time epidural catheter. Methods: SPF SD rats were divided into 3 groups with 18 in each group: the normal control group (A), mode groupl (B), and the icariin group (C). After 6h, the rats in group C were given intragastdc administration of icariin, The group A and group B were given the same amount of normal saline. Sampling was collected within 24h, 72h and 168h, respectively, 6 mice each time. Results: Under the light microscope, different levels of inflammation of patholog), were found in lung tissue in group B and C; and the Smith scores in both group B and C were higher than A (P〈0.05), But the lesion of C group were remarkably alleviated (P〈0.05). Under the transmission electron microscope, Mast Cell degranulation was obviously in group B and C. which the 24h was the most, but the group C was lighter than group B (P〈0.05). The expression of β -ttyptase in group B and C were higher than group A in these times, but it was significantly lower in group C than group B at 72 hours and 168 hours (P〈0.05), especially at 168h. Histamine content in group B and C were higher than group A (P〈0,05). There was no difference in group B and C, The same change trend of TNF- α and IL-6 content, in both group B and C were higher than group A (P〈0.05), Among the three times, they reached a peak at 24h and then declined. Conclusion: In tracheotomy intubation rat, icariin could decrease the pulmonary inflammation, The possible mechanism are icariin could inhibit the activity of mast cells.
