摘要
目的探讨Pink-1在百草枯(PQ)中毒大鼠肾损伤中的表达及意义。方法35只健康SD大鼠随机分为7组,每组5只,分别为对照组(NC组)、PQ染毒3h、6h、9h、12h、24h、48h组。用HE染色方法分别观察各组大鼠肾组织病理学变化,同时采用IHC、Western blot方法观察Pink-1蛋白表达情况。结果对照组肾脏组织结构清晰,染毒组结构清晰度下降。染毒3h即可见充血、水肿等病理变化,随时间延长而加重,肾小球亦可受累。与对照组相比,PQ组染毒9h、12h、24h、48h的Pink-1蛋白表达量上升(P<0.05)。结论Pink-1蛋白可能参与百草枯中毒肾损伤的病理机制。
Objective To study the expression and significance of Pink-1 in rats kidney induced by paraquat poisoning. Methods 35 SD mice were randomly divided averagely into: NC group, PQ 3h group, PQ 6h group, PQ 9h group, PQ 12h group, PQ 24h group, PQ 48h group. Kidney tissues were harvested and observed by HE staining. The expression of Pink-1 in rats kidney was examined by immunohistochemistry and Western blot. Results Kidney tissue structure in the control group was clear, the legibility in PQ exposure group was decreased. The hyperemia, edema and other pathological changes were observed in kidney of PQ poisoning 3h group, and more serious as time elongeted. Compared with the control group, the expression level of Pink-1 protein was increased in PQ 9h group, PQ 12h group, PQ 24h group, PQ 48h group(P〈0.05). Conclusion Pink-1 might be involved in the pathogenesis of PQ.
