摘要
目的研究白藜芦醇促骨肉瘤细胞凋亡的机制。方法 MTT法测MG-63细胞增殖。流式细胞术、TUNEL法检测白藜芦醇对骨肉瘤细胞的凋亡。Western blot法测MG-63细胞中各蛋白表达。结果相比对照组,不同浓度的白藜芦醇处理MG-63细胞的抑制率明显增加,白藜芦醇能上调p53、Bak和Bax表达增加(P<0.01),Caspase-3(p19)蛋白表达随浓度增加有递增趋势,实验组与对照组差异有统计学意义(P<0.05)。结论白藜芦醇能上调p53蛋白,并且激活Bax和Bak,抑制Bcl-2蛋白,激活Caspase-3(p19)蛋白,导致骨肉瘤细胞的凋亡。
Objective To study the mechanism of resveratrol in the apoptosis of osteosarcoma cells. Methods The proliferation of MG-63 cells was measured by MTT method. The effect of resveratrol on the apopto- sis of osteosareoma cells was detected by flow cytometry and TUNEL test. The expression of each protein in MG-63 cells was measured by Western blot. Results Compared with the control group, the MG-63 cells treated with resveratrol at different concentrations were all significantly inhibited and resveratrol upregulated the expressons of p53, Bak and Ban (P 〈 0.01 ) , and the caspase-3 (p19) expression in the experimental groups increased more significantly with the decrease of resveratrol concentrations, as compared with the control group (P 〈 0.05). Conclusion Resveratrol responds to apoptosis of osteosarcoma cells by increasing p53 protein, activating Bax, Bak, easpase-3 (P19) protein, and inhibiting Bel-2 protein.
