槐杞黄颗粒对MRL/Lpr狼疮小鼠肾损伤治疗作用的影响 被引量:12

Effect of Huaiqihuang Granule on MRL/Lpr Lupus Erythematosus Mice with Renal Injury

摘要 目的研究槐杞黄颗粒对MRL/Lpr狼疮小鼠肾损伤病理进程发展的影响。方法正常饲养C57BL/6小鼠8只为正常组。8~10周龄MRL/Lpr狼疮小鼠24只随机分为模型组、槐杞黄颗粒组(HQH组)、地塞米松组(DXM组),每组8只。MRL/Lpr狼疮小鼠12周龄左右开始发病,HQH组与DXM组分别给予槐杞黄颗粒(4 g/kg)及地塞米松(2.5 mg/kg)灌胃治疗,每天1次,共治疗15天。检测各组小鼠体重,24 h尿蛋白、白蛋白、SCr、BUN及抗核抗体(ANA)、抗双链DNA(ds DNA)含量水平,HE染色观察肾脏组织病理,免疫荧光发检测IgG及C3。结果与正常组比较,模型组体重增长差值减少,ANA、ds DNA、白蛋白、SCr、BUN水平升高(P<0.01)。与模型组比较,DXM组及HQH组体重差值增加(P<0.01),ANA、ds DNA、白蛋白、SCr、BUN水平下降(P<0.01)。与治疗前比较,治疗后DXM组及HQH组24 h尿蛋白量明显下降(P<0.05,P<0.01)。HE染色可见模型组肾小球细胞增生,且肾小球毛细血管壁有明显损伤,肾小球和肾间质炎细胞浸润,同时肾小管肿胀明显;DXM组和HQH组炎性细胞浸润明显减少,肾小管肿胀有所减轻。免疫荧光检测显示与正常组小鼠比较,模型组小鼠肾脏中IgG表达上升,C3的表达下降;DXM组及HQH组小鼠肾脏中IgG表达下降,C3表达上升。结论槐杞黄颗粒有改善MRL/Lpr狼疮小鼠肾脏病理的作用。 Objective To explore the effect of Huaiqihuang Granule on the pathological process of renal injury in MRL/Lpr lupus mice. Methods Eight C57 BL/6 mice were used as the normal group. Twenty-four 8-10-week-old MRL/Lpr lupus mice were randomly divided into three groups,i. e. model group,Huaiqihuang granule group( HQH group),dexamethasone group( DXM group),8 in each group. MRL/Lpr lupus mice began to sicken at about 12 weeks old. The mice in the HQH group were treated by Huaiqihuang Granule( 4 g/kg) and the mice in the DXM group were treated by dexamethasone( 2. 5 mg/kg),once a day,for 15 days. Body weight,24 h urinary protein,albumin,SCr,BUN,antinuclear antibody( ANA) and double strand DNA( ds DNA) levels were detected in each group. HE staining was used to observe renal histopathology,and immunofluorescence was used to detect IgG and C3. Results Compared with the normal group,the difference in weight gain of the model group decreased,and the levels of ANA,ds DNA,albumin,SCr and BUN increased( P〈0. 01). Compared with the model group,the weight gain of the DXM group and the HQH group increased(P〈0. 01),and the ANA,ds DNA,albumin,SCr and BUN decreased(P〈0. 01). The amount of 24 h urine protein in the DXM and HQH group decreased significantly after treatment(P〈0. 05,P〈0. 01). HE staining showed glomerular mesangial cells proliferation in model group,glomerular capillary wall was obviously damaged,glomerular and interstitial inflammatory cells infiltrated,and renal tubular swelling could be seen clearly. In the DXM and HQH group,inflammatory cell infiltration decreased and renal tubular swelling decreased. Compared with normal group,immunofluorescence assay showed that the expression of IgG increased and the expression of C3 decreased in the model group,but the expression of IgG and the expression of C3 increased in DXM and HQH groups. Conclusion Huaiqihuang granule can improve the renal pathology of MRL/Lpr lupus mice.
