摘要
目的研究2,5-己二酮对大鼠卵巢颗粒细胞凋亡及干细胞因子(SCF)表达的影响。方法 21日龄清洁级雌性SD大鼠10只,提取卵巢颗粒细胞混合后,均匀接种到六孔板培养,设立1个对照组和3个实验组,培养24h后分别加入浓度为0、20、40、60mmol/L的2,5-己二酮染毒24h,Hoechst 33258荧光染色计算细胞凋亡率,实时荧光定量PCR检测SCF、Bax、Bcl-2基因mRNA相对表达量。结果各染毒剂量组卵巢颗粒细胞凋亡率增高,差异有统计学意义(F=221.23,P<0.05)。SCF基因mRNA的相对表达量在40、60mmol/L剂量组呈下降趋势,与对照组比较差异有统计学意义(P<0.05)。Bcl-2基因mRNA的相对表达量随染毒剂量增加呈下降趋势,各剂量组与对照组比较差异有统计学意义(P<0.05)。Bax基因mRNA的表达量和Bax/Bcl-2比值随着染毒剂量增加呈上升趋势,各剂量组与对照组相比差异有统计学意义(P<0.05)。结论2,5-己二酮染毒可致卵巢颗粒细胞凋亡率增高,抑制SCF基因表达,SCF/c-Kit信号通路可能参与此过程。
Objective To observe the effect of 2,5-hexanedione(2,5-HD)on the apoptosis of ovarian granulosa cells and the expression of stem cell factor(SCF)in rats. Methods Ovarian granulosa cells were collected from 10 female 21 days old Wistar rats and incubated in vitro for 24 hours.The cells were randomly divided into four groups and exposed to 0,20,40 and 60 mmol/L 2,5-HD for another 24 hours.The rate of cell apoptosis was determined using Hoechst 33 258 fluorescent dye and real-time PCR was used for mRNA expressions of SCF,Bax and Bcl-2 gene. Results The apoptosis rates of ovarian granulosa cells in all three 2,5-HD dose groups were higher than that of the control group(F=221.23,P〈0.05).Compared with the control group,the relative expression of both SCF and Bcl-2 decreased significantly,especially in 40 and 60 mmol/L2,5-HD dose groups(P〈0.05).However,the relative expression of Bax and the ratio of Bax/Bcl-2 increased with the increase exposure dose of 2,5-HD,the differences were significant compared with the control group(P〈0.05). Conclusions 2,5-HD exposure may increase the apoptosis rate of ovarian granulosa cells in rats possibally by inhabiting the expression of SCF gene through SCF/c-Kit signaling pathway.
