摘要
目的:优选化扁汤的提取工艺,为该制剂的工业化生产提供参考。方法:以绿原酸、黄芩苷、牛蒡苷和连翘苷的总含量及干膏得率的综合评分为指标,通过L9(34)正交试验考察提取次数、提取时间和加水量对提取效果的影响。采用多波长切换HPLC法同时测定绿原酸、黄芩苷、牛蒡苷和连翘苷的含量,以流动相乙腈-0.2%磷酸溶液梯度洗脱,DAD检测器,检测波长为327 nm(0~37 min,检测绿原酸),278 nm(37~41.3 min,检测黄芩苷),228 nm(41.3~50 min,检测连翘苷和牛蒡苷)。流速:1.0 m L·min-1;柱温:35℃。结果:最佳水提工艺为加8倍量水煎煮3次,每次1 h;绿原酸、黄芩苷、牛蒡苷和连翘苷的总提取量为3.50 mg/m L,干膏得率为27.09%。结论:优选的提取工艺稳定,重复性好,可为化扁汤的工业化生产提供参考。
Objective: To optimize the extraction process of Huabian Oral Liquid and provide the reference for the industrialized production. Methods: With composite scores of extraction amounts of chlorogenic acid,baicalin,phillyrin,arctiin and dry paste yield as the comprehensive evaluation indexes,orthogonal test of L9( 34) was adopted to investigate effects of the amount of wate,decocting time and times on extraction process. Amulti wavelength switching HPLC method was used for simultaneous determination of chlorogenic acid,baicalin,arctiin and phillyrin contents in mobile phase acetonitril-0. 2 phosphoric acid solution with gradient elution and DAD detector. The detection wavelength was 327 nm( 0-37 min for chlorogenic acid),278 nm( 37-41. 3 min for baicalin) 228 nm( 41. 3-50 min for phillyrin and arctiin),flow rate at 1. 0 m L· min-1. The column temperature was at35℃. Results: Water extraction technology was as follows: added 8-fold water,each time for 1 hour. The total extracting amounts of chlorogenic acid,baicalin,phillyrin and arctiin were 3. 50 mg/m L and the yield of dry extract was 27. 09%. Conclusion: The optimized extraction process is stable and reproducible,and it can be used as a reference for the industrialized production.
