针刺对宫内窘迫大鼠缺氧缺血性脑损伤作用机制初探 被引量:5

Study on Mechanism of Acupuncture on Hypoxic-Ischemic Brain Damage in Models of Intrauterine Distress Rats

摘要 目的:观察"靳三针疗法"中的"头针穴组"对宫内窘迫导致的HIBD新生大鼠脑皮质中线粒体氧化应激及能量代谢的影响。方法:给予临产前孕鼠异氟烷吸入麻醉和夹闭双侧子宫动脉10 min的延迟剖宫产术造缺氧缺血性脑损伤(HIBD)新生大鼠模型。正常分娩的新生大鼠为正常组;造模成功的HIBD新生大鼠按照随机数字表法分为模型组和针刺组,每组6只幼鼠。针刺干预于大鼠14天鼠龄时进行,每天针刺1次,共干预14 d。针刺干预结束后取各组大鼠的全脑进行指标检测。采用流式细胞术检测大鼠脑皮质中ROS的表达;采用高效液相色谱法检测大鼠脑皮质中ATP/ADP比值。结果:(1)各组大鼠脑皮质重量比较:与正常组相比,针刺组脑皮质重量差异不显著(P〉0.05),模型组脑皮质重量显著下降(P〈0.05);与模型组比较,针刺组脑皮质重量明显优于模型组,差异有统计学意义(P〈0.05)。(2)各组ROS平均荧光表达比较:模型组ROS平均荧光表达明显高于正常组;与模型组相比,针刺组大鼠的ROS表达明显降低,差异具有显著性(P〈0.05)。(3)比较各组的ATP/ADP比值:与正常组相比,模型组与针刺组均低于正常组且差异有显著性(P〈0.05),与模型组相比,针刺组高于模型组,差异有统计学意义(P〈0.05)。结论:(1)针刺靳三针头针穴组能有效促进缺氧缺血性脑损伤HIBD模型大鼠脑皮质重量增长;(2)针刺组中ROS与ATP/ADP比值表达成负相关趋势,提示针刺靳三针头针穴组促进缺氧缺血性脑损伤HIBD模型大鼠脑功能修复作用机制可能与针刺靳三针头针穴组能有效降低HIBD模型大鼠脑组织中的氧化应激损伤,并能提高HIBD模型大鼠脑组织线粒体能量代谢有关。 Objective: To observe the effect of Jin’s three-needle therapy on mitochondrial oxidative stress and energy metabolism in the cerebral cortex of neonatal rats with hypoxic-ischemic brain damage. Methods: Pregnant rats which were going to give birth to their babies were given an isoflurane inhalation anesthesia to receive a delay caesarean delivery with clipping bilateral uterine artery for 10 minute to create neonatal rat models with hypoxic-ischemic brain damage( HIBD). The rats were delivered without intervention were in the normal group and the anoxic rats were divided into model and acupuncture groups randomly,with6 rats in each group. Rats in acupuncture group received the acupuncture intervention for 2 weeks when they were 14 days old. All rats were killed by beheading on day 28 with whole brain tissue for detection. Fluorescent expression of ROS and ratio of ATP/ADP in cerebral cortex were detected by flow cytometry and reversed-Phase high-Performance liquid chromatography respectively. Results:( 1) Compared with normal group,the cerebral cortex weight of acupuncture group was not significant( P 〉 0. 05)and the cerebral cortex weight of model group decreased markedly( P 〈 0. 05). In comparison with model group,the cerebral cortex weight of acupuncture group was better than that of model group,which was statistically significant( P 〈 0. 05).( 2) Fluorescent expression of ROS in model group was significantly higher than that of the normal group and acupuncture group( P 〈 0. 05),while the ratio of ATP/ADP in model group was the lowest,comparing with taht of the normal and acupuncture groups( P 〈0. 05). In comparison with model group,fluorescent expression of ROS in acupuncture group was lower than that of the model group while the ratio of ATP/ADP was higher than that of the model group,both of which were significant( P 〈 0. 05). Conclusion:( 1) Acupuncture could promote growth of cerebral cortex weight in rats with hypoxic-ischemic brain damage effectively.( 2) The ratio of ROS to ATP/ADP in acupuncture group was expressed as a negative correlation trend,which suggested that acupuncture of Jin’s three-needles could promote the brain function to repair mechanism of HIBD model rats with hypoxic-ischemic brain damage which might be related to acupuncture function of reducing oxidative stress injury but improving mitochondrial energy metabolism of brain tissues of HIBD model rats effectively.
