绿色高效液相色谱法测定饮料中4种合成着色剂 被引量:6

Simultaneous determination of 4 synthetic colorants in beverage by green high performance liquid chromatography

摘要 目的建立以含表面活性剂的缓冲溶液为流动相的高效液相色谱法同时测定饮料中柠檬黄、苋菜红、胭脂红和日落黄。方法为实践绿色分析理念,分析过程中不使用有机溶剂。本研究优化了流动相中表面活性剂的浓度、磷酸盐缓冲溶液的浓度和p H值。样品经处理后,以C18柱为分离柱,0.5%Triton X-100-50 mmol/L磷酸盐缓冲溶液(p H=7.0)为流动相,等度洗脱,多波长检测,标准曲线法定量。结果 4种合成着色剂的线性相关系数均>0.999,回收率为97.8%~102%,RSD为0.5%~2.0%,最低检出限为0.1 mg/kg^1.0 mg/kg。本法与国家标准方法 GB 5009.35—2016比较,实际样品测定结果差异无统计学意义。所检市售饮料中4种合成着色剂都有添加,含量均小于食品国家安全标准限值。结论该法不使用有机溶剂,绿色环保;方法简便,快速,准确,灵敏,适用于饮料中4种合成着色剂的定量分析。 Objective A method was developed for the simultaneous determination of lemon yellow,amaranth,carmine and sunset yellow in beverage by green high performance liquid chromatography(HPLC)with the buffer solution containing surfactant as mobile phase.Methods In order to practice the concept of green analysis,organic solvents was not used in the analysis.The concentration of surfactant,the concentration of phosphate buffer solution and p H value were optimized.The samples were pretreated,and 4 colorants were separated by C18 column using 50 mmol/L phosphate buffer solution containing 0.5% Triton X-100(p H = 7.0)as the mobile phase,and then detected by multi-wavelengths,with quantification based on standard curves.Results The linear correlation coefficients(r)were greater than 0.999.The recoveries were within 97.8%-102%and the RSDs were within 0.5%-2.0%.The detection limits of 4 colorants were within 0.1 mg/kg-1.0 mg/kg.There was no statistical significance on the difference between the results of the method for actual samples with those by GB 5009.35—2016.Four synthetic colors were added in the seized beverages,and the contents were all less than the national food safety standards.Conclusion The established method is green due to without using organic solvent.The method is simple,rapid,accurate and sensitive,and it is suitable for quantitative analysis of four synthetic colorants in beverage.
