摘要
目的对影响狗肾传代细胞(MDCK细胞)分离流感病毒阳性率的相关因素进行单因素分析。方法选择2013年-2016年南阳市哨点医院送检流感样病例(ILI)咽拭子中PCR检测阳性的标本,采用MDCK细胞培养法进行流感病毒的分离培养,并对咽拭子标本从采样、核酸检测和细胞分离等环节中的一些因素进行卡方检验和分析。结果不同监测年度、新甲型H1N1流感病毒核酸检测Ct值大小对病毒分离结果影响差异有统计学意义(P<0.05);不同流感病毒型别(或系)的MDCK细胞一代分离培养阳性率差异有统计学意义(P<0.05)。结论不同的监测年度,病毒分离阳性率也不同;新甲型H1N1流感病毒核酸检测Ct值大小影响其病毒分离阳性率,Ct值越小,病毒分离阳性率越高;不同流感病毒型别MDCK细胞一代病毒分离阳性率不同,季甲型H3流感病毒MDCK细胞分离培养第一代阳性率明显低于B型和新甲型H1N1流感病毒。
Objective Single factor analysis is carried out for the relevant factors affecting the positive rate of cell influenza virus isolated Madin-Darby canine kidney(MDCK).Methods The samples with positive PCR test result of the throat swab specimens from the influenza-like illness(ILI)submitted to Nanyang sentinel hospitals during 2013-2016 were collected for isolation and cultivation of influenza virus by MDCK cell culture method,and chi-square test and analysis were carried out for some factors in the links,such as sampling,nucleic acid detection and cell isolation of the throat swab samples.Results The Ct value differences of nucleic acid detected in different monitoring years and H1 N1 influenza virus had statistical significance on the results of virus isolation(P〈0.05);the positive rate difference in the isolation and culture of MDCK cells in different influenza virus types(or subtypes)was statistically significant(P〈0.05).Conclusion For different monitoring years,the positive rate for isolation of virus is also different;the Ct value for the nucleic acid detection of the new A H1 N1 influenza virus affects its positive rate of virus isolation,so the smaller Ct value is,and the higher positive rate for virus isolation will be.The positive rate for the isolation of the first generation of the virus by MDCK cell is different for different types of influenza virus.The positive rate for isolation of the first generation of A H3 influenza virus by MDCK cell is significantly lower than that of type B and new A H1 N1 influenza virus.
