摘要
目的通过检测妊娠高血压模型大鼠胎盘中可溶性血管内皮生长因子受体-1(s Flt-1)和胎盘生长因子(PLGF)的含量变化,初步探讨黄芪注射液对妊娠高血压大鼠的治疗作用。方法将稳定的妊娠高血压动物模型鼠随机分为5组:分别为模型对照组、黄芪注射液低剂量组、黄芪注射液中剂量组、黄芪注射液高剂量组、硫酸镁组,每组6只,同时选取健康的妊娠雌鼠6只作为空白对照组。给予腹腔注射低、中、高剂量黄芪注射液,并以空白组、模型组和硫酸镁组作为对照,观察治疗前后孕鼠血压,尿蛋白水平;于妊娠第21日剖腹取胎盘,提取胎盘组织中核糖核酸(RNA),采用反转录聚合酶链反应(RT-PCR)法分别测定上述2种因子含量的变化。结果黄芪低、中、高组大鼠与模型组大鼠比较,胎盘中s Flt-1 m RNA的表达明显降低,PLGF m RNA的表达明显增高,差异有统计学意义(均P<0.05)。结论黄芪注射液干预妊娠高血压模型鼠后,大鼠胎盘s Flt-1的表达含量降低,PLGF的表达含量升高。
Objective To determine the changes of placenta soluble vascular endothelial growth factor receptor-1（s Flt-1） and placental growth factor（PLGF） contents in the rat models of gestational hypertension, and to preliminarily determine the effect of Astragalus injection on gestational hypertension in rats. Methods The stable rat models of gestational hypertension were randomly divided into five groups： the model control group, low-dose Astragalus injection group, medium-dose Astragalus injection group, high-dose Astragalus injection group, and magnesium sulfate group（n=6 each）. At the same time, 6 healthy pregnant rats were included as the blank control group. The Astragalus injection groups were injected intraperitoneally with low, medium and high-doses Astragalus injection, and the blank group, model group and magnesium sulfate group were taken as the controls. The blood pressure and urinary protein levels in the pregnant rats were determined before and after the treatment. At 21 d after the pregnancy,the removal of placenta was performed by caesarean section to extract the ribose nucleic acid（RNA） from the placenta tissues. The reverse transcription polymerase chain reaction（RT-PCR） was used to determine the changes of the two contents. Results Compared with the model group, the expression of s Flt-1 m RNA significantly decreased,whereas the expression of PLGF m RNA significantly increased in the placenta of low, medium and high-doses Astragalus injection groups（all P〈0.05）. Conclusion After the intervention of Astragalus injection in the rat models of gestational hypertension, the s Flt-1 expression level decreases and the PLGF expression level increases in the rat placenta.
