摘要
目的体外研究IL-17抗体对大鼠破骨细胞功能的影响。方法分离并培养大鼠破骨细胞,体外检测IL-17抗体对大鼠破骨细胞功能的影响,利用Realtime-PCR、ELISA、Western-blot检测经IL-17刺激及IL-17抗体阻断后大鼠破骨细胞中cathepsin K、MMP9表达情况,所得数据经SPSS 13.0进行统计学分析。结果大鼠破骨细胞经IL-17刺激后,能够显著提高cathepsin K、MMP9的表达水平,经IL-17抗体阻断后cathepsin K、MMP9的水平均恢复到接近对照组水平。结论 IL-17通过上调cathepsin K、MMP9的表达量,从而增强破骨细胞降解骨基质的能力。
Objective The effect of il-17 antibody on osteoclast function in rats was studied in vitro. Methods Separate and culture rat osteoclasts, the expression of cathepsin K and MMP9 was measured by Realtime-PCR、ELISA、Western-Blot to test the effect of IL-17 and IL-17 antibody on osteoclast cells, the data were statistically analyzed by spss13.0. Results Compared to the control group, the expression levels of cathepsin K and MMP9 of osteoclasts were increased significantly with the stimulation of IL-17. The effect of IL-17 was inhibited by IL-17 antibody. Conclusion We observed that IL-17 stimulated the expression of cathepsin K、MMP9 in osteoclast cells to enhance th e ability of osteoclasts degradation of bone matrix.
