摘要
目的研究白芍提取物对小鼠抗抑郁及抗炎作用的影响。方法雄性昆明小鼠随机分为正常对照组、模型组、白芍提取物高剂量组[255 mg/(kg·d)]和白芍提取物低剂量组[95 mg/(kg·d)]、盐酸氟西汀组[3.033 mg/(kg·d)],其中利血平诱导的急性抑郁症实验,分组及给药方法同上,除正常对照组外,其余均腹腔注射利血平[5mg/(kg·d)],连续给药7 d,利用小鼠悬尾实验(TST)和小鼠强迫游泳实验(FST)评价药效;使用利血平拮抗实验评价对于中枢单胺神经功能的影响。用二甲苯致耳肿胀方法评价抗炎效果。结果 FST和TST中,白芍提取物高剂量组与模型组比较,相对不动时间明显缩短(P<0.05);在利血平拮抗实验中,白芍提取物低剂量组可拮抗利血平诱导的小鼠眼睑下垂、肛温下降和运动不能(P<0.05)。抗炎实验结果显示白芍提取物低剂量组能显著改善小鼠耳肿胀炎症症状(P<0.05)。结论白芍提取物对小鼠有抗抑郁及抗炎作用,可能通过影响中枢单胺神经功能对抑郁及炎症进行调节。
Objective To study the effects of radix paeoniae alba extract on anti-depression and anti-inflammation. Methods The male Kunming mice were randomized into a normal control group,a model group,a high-dose 255 mg/( kg·d) radix paeoniae alba extract group,a low-dose 95 mg/( kg·d) radix paeoniae alba extract group and fluoxertine hydrochloride 3. 033 mg/( kg·d) group. In the acute depression experiment induced with reserpine,the group division and medications were same as the above. Except the mice in the normal control group,the intraperitoneal injection with resperpine 5 mg/( kg·d) was given,continuously for 7 days. The tail suspension test( TST) and the forced swimming test( FST) were used to evaluate the pharmaceutical effects. The reserpine antagonistic test was adopted to evaluate the effects on the central monoamine neural function. With the ear swelling method induced with xylene,the effects on anti-inflammation were evaluated. Results In FST and TST,in comparison between the high-dose radix paeoniae alba extract group and the model group,the relative immobility time was shortened remarkably( P〈0. 05). In the reserpine antagonism experiment,in the low-dos group,the eyelid potosis,reduced annual temperature and akinesis were induced( P〈0. 05). The anti-inflammation test showed that the low-dose radix paeoniae alba extract relieved significantly the ear swelling inflammatory symptoms in the mice. Conclusion Radix paeoniae alba extract acts on anti-depression and anti-inflammation in the mice,possibly through the influences of central monoamine neural function on the regulation of depression and inflammation.
