摘要
目的观察依普利酮对急性心肌梗死(AMI)大鼠心肌炎症反应、心脏功能的影响。方法将结扎左冠状动脉致AMI大鼠模型随机分为AMI组(n=11)和依普利酮治疗组(AMI—E,n=11),并设立假手术(Sham,n=14)组。在依普利酮(30mg·kg^-1·d^-1)治疗4周后,用超声心动图和血流动力学指标评价心脏功能;心肌Masson染色法测定心肌胶原容积分数(CVF)和血管周围胶原面积(PVCA),碱水解法测定羟脯氨酸(Hyp)含量;酶联免疫吸附法测定心肌组织中肿瘤坏死因子-α(TNF—α),白介素-1β(IL-1β)、白介素-6(IL-6)。结果①AMI组大鼠心肌CVF、PVCA、Hyp较Sham组显著增加(P〈0.01);AMI—E组大鼠心肌CVF、PVCA、Hyp较AMI组均显著减低(P〈0.05)。②AMI组大鼠心肌组织中TNF—d(10.7±3.2)pg/g、IL—1β(14.5±3.5)Pg/g、IL-6(21.1±8.0)Pg/g表达较Sham组[(5.6±1.1)pg/g、(8.2±2.1)pg/g、(9.8±3.1)pg/g]显著增加(P〈0.01);AMI—E组TNF—α(7.8±2.6)Pg/g、IL-1β(11.3±3.2)Pg/g、IL-6(14.7±4.7)Pg/g表达较AMI组显著减轻(P〈0.05),并伴随心脏功能的改善(P〈0.05)。结论依普利酮改善AMI大鼠心脏功能,可能与抑制心肌炎症反应、减轻心肌纤维化有关。
Objective To investigate the effect of Eplerenone on cardiac inflammation in rats with acute myocardial infarction. Methods The rat model of AMI was established by ligating the left coronary artery in male SD rats. The survived rats were randomly divided to AMI group (n= 11 ), Eplerenone (AMI-E, n= 11 ) group, and sham-operated group(n=14). After four weeks, cardiac function was assessed by eehocardiogram and hemodynam- ie. The collagen volume fraction(CVF) and perivascular collagen area(PVCA ) of cardiac tissues were calculated by Masson's Trichrome. Alkaline hydrolysis method was used to detect the concentration of hydroxyproline (Hyp) in the myoeardium of left ventricle. The cardiac contents of tumor necrosis faetor-α (TNF-α), interleukin-1β(IL- 1β), interleukin-6(IL-6) were determined by ELISA. Results (1)The expression of CVF, PVCA and Hyp in the cardiac tissues were increased dramatically in AMI group compared with sham-operated group(P〈0.01 ). After four weeks treatment with Eplerenone, the expression of CVF ,PVCA and Hyp continuous decreased than those in AMI group(P〈0.05). (2)In AMI group, the cardiac contents of TNF-α(10.7±3.2)pg/g, IL-1β(14.5±3.5)pg/g, IL-6 (21.1±8.0)pg/g were significantly higher than those in sham-operated group[ (5.6±1.1)pg/g, (8.2±2.1)pg/g, (9.8± 3.1 )pg/g ] ( P〈0.01 ). After four weeks treatment with Eplerenane, TNF-ct ( 7.8 ±2.6 )pg/g, IL- 113 ( 11.3 ±3.2 )pg/g, IL-6 (14.7±4.7)pg/g were reduced obviously compared with AMI group. At the same time, cardiac function was improved in AMI-E group (P〈0.05). Conclusion Eplerenone can improved cardiac function following AMI, which partly contributes to attenuating cardiac inflammation and cardiac fibrosis.
