摘要
目的:观察保肝复方三味干姜散对菌群失调联合肝损伤动物模型的影响,为保肝作用与肠道菌关系做初步探索。方法:菌群失调联合肝损伤动物模型,在连续3天灌胃克林霉素后,给予三味干姜散7天,末次给药后皮下注射CCl_4,测定肝功能酶ALT、AST;HE染色法观察肝肠组织病理学变化;16 Sr DNA测序法检测肠道菌群主要菌属的变化情况,同时设定肝损伤模型进行对比。结果:菌群失调联合肝损伤模型显示,三味干姜散作用后能显著降低肝损伤模型及联合模型ALT、AST水平;减轻肝脏损害程度,改善肠粘膜水肿情况;拟杆菌门、厚壁菌门水平升高,变形菌门水平显著下降,Parabacteroide水平显著下降,螺杆菌属的水平下降,支原体属水平下降。结论:三味干姜散能改善联合模型的肝损伤程度及菌群失调水平,说明三味干姜散可能具有保肝和调节肠道菌群的双重作用。
Objective: To observe the effect of Sanwei Ganjang Powder(SWGJ) on the animal model of dysbacteriosis combined liver injury,to make a preliminary exploration of the relationship between liver and intestinal bacteria. Methods: The combined model was established by orally giving Clindamycin for 3 days,and giving SWGJ for 7 days,injecting CCl_4 oil solution subcutaneously. ALT and AST were detected,pathological changes of livers and ileum tissues and the main intestinal bacteria change were observed on the combined model. Results: The result of combined model showed that,SWGJ significantly reduced levels of ALT and AST(P〈0. 01 or P〈0. 05). SWGJ improved the liver damage. SWG improved the intestinal mucosa edema in the combined model. SWGJ increased the phylum level species abundance of Bacteroidetes and Firmicutes,significantly decreased the level of Proteobacteria(P〈0. 05),SWGJ significantly decreased the genus species abundance of Parabacteroide(P〈0. 05),decreased the level of Helicobacter and Mycoplasma. Conclusion: SWGJ can improve the liver injury degree and dysbacteriosis level in the combined model,illustrating that the regulatory effect of SWGJ on the combined model is probably concurrently protecting the liver and regulating the intestinal flora.
