摘要
本文通过对饲料中油脂提取方法进行优化,建立了检测饲料中油脂过氧化值的有效方法。优化后的过氧化值测定方法为:将TBHQ(m(TBHQ)=饲料重×饲料含油率×200 mg/kg)添加到饲料中搅拌均匀,称取50 g含TBHQ的饲料到500 mL圆底烧瓶中,加入150 mL[料液比(g/mL)=1∶3]正己烷。在30℃搅拌下回流浸取0.5 h。反应结束后,在4500 r/min下离心5 min,然后旋蒸,旋蒸脱溶后按GB/T 5538-2005中方法测定过氧化值。结果表明,过氧化值为3~20 mmol/kg,线性关系良好,线性相关系数为0.9981,多次实验平均过氧化值(PV)回收率为95.56%~106.83%,且不受饲料含油率影响。
In this paper,the method of oil extraction in feed was optimized,and an effective method for the determination of lipid peroxidation in feed was established.In this paper,the detection method of peroxide value for oil in feed was optimized as follows:TBHQ(TBHQ=200 mg/kg oil in feed) was added to the feeds,and 50 g of feeds containing TBHQ and 150 mL of n-hexane(g/mL =1∶3) were added to a 500 mL round-bottomed flask in a magnetic stirrer for 0.5 h.After that,the supernatant was centrifuged at 4500 r/min for 5 min,evaporated by rotary evaporation apparatus with water bath at 30 ℃,80 ~ 100 r/min and 20 ~ 30 k Pa,after that the peroxide value of oil was detected according to the standard method.The results showed that the linear correlation coefficient was over 0.9981 and the average peroxide value(PV) recoveries were between 95.56%and 106.83%.The relatire standard deviation(RSD) ranged from 0.46% to 3.63% in the range of 3 ~ 20 mmol/kg and not be affected by oil content in feed.
