摘要
目的探讨褪黑素对大鼠糖尿病肾病肾脏的影响。方法将24只大鼠只随机分为正常(NC)组、糖尿病肾病(DN)组和褪黑素(Mel)组。DN组、Mel组采用一次性腹腔注射STZ30mg/kg,给予高脂饮食;NC组注射等体积的柠檬酸缓冲液,给予普通饲料。禁食6小时后,Mel组、DN组给予褪黑素20mg/kg/d,NC组给予等量含有2%乙醇的生理盐水溶液灌胃,连续6周后,检测FBG;心脏采血检测血BUN、Scr、24h UP;Western blot法检测caspase-3蛋白;肾组织匀浆检测MDA、SOD、GSH-Px;组织切片观察大鼠肾组织形态学变化,TUNEL检测细胞凋亡指数(AI)。结果与NC组比较,DN组FBG、BUN、Scr和24h UP显著升高(P<0.05),AI增高(P<0.05);形态学显示肾小管上皮细胞水肿,内皮细胞和系膜细胞增生显著,基底膜增厚,肾小球纤维化,间质纤维组织增生;与DN组比较,Mel组BUN、Scr、24h UP均明显降低(P<0.05),肾组织SOD、GSH-Px活性明显升高(P<0.05),MDA含量明显降低(P<0.05),AI显著降低(P<0.05);形态学显示肾小管水肿较明显减轻,纤维化有明显的改善。结论褪黑素可通过减轻氧化应激损伤和减少肾脏细胞凋亡,对大鼠DN肾脏具有一定的保护作用。
Objective To explore the effect of melatonin on kidney of rats with diabetic nephropathy(DN).Methods24 rats were randomly divided into 3 groups:normal control(NC) group,diabetic nephropathy(DN) group and melatonin(Mel) group; intraperitoneal injection of STZ 30 mg/kg and high-fat diet were made to rats in DN group and in Mel group while citrate buffer of same amount and normal diet were made to rats in NC group; after fasting for 6 hours,rats in Mel group and DN group were given melationin 20 mg/kg/d while rats in NC group were gavaged normal saline solution containing 2% ethanol; the operation kept from 6 weeks and then FBG was detected; blood was drawn from the heart for the detection of BUN,Scr and 24-hour UP; Western blot was applied in detecting caspase-3 protein; MDA,SOD and GSH-Px in kidney homogenate were detected; themorphological changes in rat kidney were observed with tissue section and TUNEL was applied in detecting the cell apoptosis index(AI).Results The increases of FBG,BUN,Scr and 24-hou UP in DN group were much greater than those in NC group(P〈0.05) and AI increased much more as well(P〈0.05); morphology indicated hydrops of renal tubular epithelial cell,endothelial and mesangial cell proliferation,basement membrane thickening,glomerular fibrosis and the hyperplasia of the interstitial fiber;compared with DN group,BUN,Scr and 24-hou UP in Mel group decreased much more(P〈0.05),the acitivity of SOD and GSH-Px of renal tissues increased much more(P〈0.05),the content of MDA and AI dropped down more obviously(P〈0.05); morphology indicated an obvious improved hydrops of renal tubular epithelial cell and an obvious improvement in fibrillation.Conclusions Melatonin is of certain protective effect on the kidney of DN rats by reducing oxidative stress injury and decreasing renal cell apoptosis.
