摘要
目的:研究葱白提取物对急性心肌梗死后心力衰竭(心衰)大鼠的保护作用。方法:将40只雄性SD大鼠随机分为假手术组和模型组,模型组结扎左冠状动脉,假手术组只穿针不结扎。造模成功后,将模型组大鼠随机分成空白模型组、培哚普利组及葱白提取物组。造模1周后,假手术组和空白模型组以0.9%氯化钠溶液灌胃,其余两组以同等剂量相应药物灌胃;连续给药12周后取左心室做HE染色、Masson染色并测定血清中B型脑钠肽(BNP)、血管紧张素Ⅱ(AngⅡ)、转化生长因子β1(TGF-β1)含量,用实时荧光定量分析技术(RT-qPCR)和免疫蛋白印记法(Western blot)检测各组大鼠心肌组织中ColⅠmRNA、ColⅢmRNA及ColⅠ蛋白的表达。结果:组织病理学显示培哚普利组及葱白提取物组较空白模型组心肌纤维化程度低。与假手术组比较,空白模型组、培哚普利组、葱白提取物组血清BNP含量显著升高(均P<0.01);与空白模型组比较,培哚普利组和葱白提取物组血清BNP、AngⅡ及TGF-β1含量均明显下降(均P<0.01),培哚普利组和葱白提取物组间差异无统计学意义;空白模型组、培哚普利组、葱白提取物组ColⅠmRNA、ColⅢmRNA及ColⅠ蛋白的表达均显著高于假手术组(均P<0.01),培哚普利组和葱白提取物组显著高于空白模型组(P<0.01),培哚普利组与葱白提取物组间差异无统计学意义。结论:葱白提取物具有治疗心肌梗死后心力衰竭大鼠的作用,降低ColⅠ/ColⅢ比例,延缓心肌纤维化,其作用机制可能是抑制AngⅡ-AT1-TGF-β1信号通路。
Objective:To study the protective effect of Fistular onion stalk extract on rats with heart failure after acute myocardial infarction(AMI).Method:Forty male SD rats were randomly divided into sham group and model group,the left coronary artery was ligated in model group,while only needled without ligation in sham group.Rats in model group were randomly divided into blank model group,perindopril group and Fistular onion stalk extract group.One week later,rats in sham group and blank model group were given normal saline,rats in other two groups were given the same dose of corresponding drugs.After 12 weeks,left ventricles were observed by HE staining and Masson staining,levels of BNP,AngⅡand TGF-β1 were determined.Levels of mRNA expression of collagenⅠand collagen Ⅲ were detected by real-time fluorescent quantitative PCR and protein levels were detected by Western blot.Result:Histological observation showed that,compared with blank model group,two drug groups had low degree of myocardial fibrosis(MF).Compared with sham group,levels of serum BNP in blank model group,perindopril group and Fistular onion stalk extract group were significantly increased(all P0.01).Compared with blank model group,levels of BNP,AngⅡand TGF-β1 were decreased obviously(all P0.01),but there was no difference between perindopril group and Fistular onion stalk extract group.Compared with sham group,expression levels of collagenⅠand collagen Ⅲ mRNA and protein were higher in blank model group,perindopril group and Fistular onion stalk extract group(all P0.01),and were higher in perindopril group and Fistular onion stalk extract group than those in blank model group(all P〈0.01),but there was no difference between perindopril group and Fistular onion stalk extract group.Conclusion:Fistular onion stalk extract can treat heart failure after AMI in rats by reducing the proportion of collagenⅠand collagenⅢ and delaying MF,the mechanism maybe relate to the inhibition of AngⅡ-AT1-TGF-β1 signal pathway.
