摘要
目的:探讨建立核酸检测实验室参考区间的可行性。方法:对两个不同厂家试剂的酶联免疫吸附法(ELISA)血液筛查乙型肝炎病毒表面抗原(HBsAg)阴性和ALT血液筛查阴性的无偿献血者血液标本,采用中山达安基因核酸检测系统进行乙型肝炎病毒核酸(HBV—DNA)8混样检测,8混样核酸检测阳性结果再进行拆分单样本核酸检测;对混样核酸检测阳性结果并且拆分单样本核酸检测结果为阳性标本进行Ct值分析,确定以Ct值为HBV—DNA阳性值的参考区间。结果:肇庆市中心血站的2017年的40020份无偿献血者血液标本,共有37份标本结果HBV—DNA阳性;Ct值为HBV—DNA阳性值的参考区间为38.26±5.586。结论:建立核酸检测实验室的阳性值参考区间,可减少核酸检测的漏检,能更有效保障血液安全。
Objective: To explore the feasibility of establishing the nucleic acid detection laboratory reference interval. Methods: Blood samples, negative in hepatitis B vires surface antigen (HBsAg) and ALTbased on two different reagents of enzyme-linked immunosorbent assay (ELISA) screening, were detected by hepatitis B virus (HBV) nucleic acid detection using Zhongshan David gene by 8 mixed sample testing. The split single sample were further detected once 8 mixed sample positive; Ct value of nucleic acid detection positive results and split single sample nucleic acid detection results were analyzed, to determine the Ct value of HBV-DNA positive reference interval. Results: A total of 37 blood samples from 40020 blood donors in Zhaoqing blood center in 2017 were found to be HBV-DNA positive. The reference interval of Ct positive value of HBV-DNA was (38.26 ±5.586) . Conclusion: Establishing the reference range of the positive value of HBV-DNA testing is benefit in reducing the leakage of HBV infection, and can ensure the safety of blood more effectively.
