摘要
目的:探讨清肺口服液治疗RSV肺炎的作用机制。方法:建立RSV感染的16HBE细胞模型,随机分为正常细胞组、空白组、模型组、清肺口服液含药血清低剂量组和高剂量组,采用Real-time RT-PCR、Western blot和免疫荧光法分别检测细胞中STAT5和IL-10的表达水平以及RSV在细胞中的相对表达。结果:与正常细胞组和空白组相比,模型组STAT5 mRNA和IL-10蛋白水平降低(P〈0.05),RSV荧光强度较高(P〈0.01);与模型组比较,含药血清组低剂量组STAT5水平升高(P〈0.05),含药血清组IL-10水平升高(P〈0.05),含药血清组RSV荧光强度降低(P〈0.01)。结论:清肺口服液对STAT5和IL-10有一定的调控作用,可能是其抗RSV感染的内在机制。
Objective: To explore the therapeutic mechanism of Qingfei Oral Liquid on RSV pneumonia. Methods:RSV infected human airway epithelial cells(16 HBE) models were randomly divided into normal cells group,blank group,model group,serum containing Qingfei Oral Liquid low and high dose groups. Real-time RT-PCR,Western blot and Immunofluorescence were used to detect the levels of STAT5,IL-10 and relative expression of RSV in the cells. Results: Compared with normal cells and blank groups,the mRNA level of STAT5 and the protein expression of IL-10 in the model group were lower(P〈0. 05);the fluorescence intensity of RSV in the model group was higher(P〈0. 01). Compared with the model group,the mRNA level of STAT5 in serum containing Qingfei Oral Liquid low dose group was increased(P〈0. 05);the protein level of IL-10 in serum containing Qingfei Oral Liquid groups were elevated(P〈0. 05) and the RSV fluorescence intensity in serum containing Qingfei Oral Liquid groups were lower(P〈0. 01).Conclusion: Qingfei Oral Liquid has a regulatory effect on STAT5 and IL-10,which may be the mechanism of its against RSV infection.
