摘要
目的探讨Rg3抑制大肠癌高侵袭细胞系Lovo/5-Fu的侵袭能力和逆转化疗耐药的机制。方法应用MTT法、穿膜实验、细胞膜流动性实验、Western blot等检测方法,检测不同浓度Rg3原液对Lovo/5-Fu逆转耐药的机制、细胞膜流动性的影响、细胞侵袭和转移能力及对转移和耐药相关蛋白nm23、caspase-8、LRP表达的影响。结果 Rg3作用后,Lovo/5-Fu细胞对氟尿嘧啶类的耐药性下降(P<0.05),细胞膜的流动性减低(P<0.05),细胞的穿膜能力减低(P<0.05),Rg3作用后caspase-8及nm23的表达明显上调,耐药相关蛋白LRP的表达无明显改变。结论 Rg3通过上调nm23的表达抑制了Lovo/5-Fu细胞的侵袭和转移能力,降低细胞膜的流动性而逆转耐药与上调caspase-8的表达相关。
Objective To explore the mechanisms of Rg3 in restraining the metastasis and reversing multidrug resistance of Lovo/5-Fu colorectal carcinoma cells. Methods MTT was applied to determine the chemosensitivity of Lovo/5-Fu to cisplatin; cell invasion assay was used to measure the invasion change of Rg3 to cells; cell membrane lipid fluidity was determined to observe the effects of Rg3 on cells,and the protein expression of nm23,caspase-8 and LRP was tested by Western blot. Results After the treatment of Rg3,Lovo/5-Fu cells had increased sensibility to 5-Fu(P〈0. 05),cell membrane lipid fluidity(P〈0. 05) as well as invasion ability(P〈0. 05) was reduced. Expressions of caspase-8 and nm23 were markedly increased,but the resistance related protein LRP had no significant change. Conclusion Rg3 can inhibit the invasion and metastasis of Lovo/5-Fu cells by upregulating the expression of nm23,and decrease the cell membrane lipid fluidity to reverse the drug resistance which is closely associated with the enhanced caspase-8.
