上皮细胞膜蛋白2在甲状腺乳头状癌中的表达及其对细胞侵袭迁移能力的影响 被引量:1

摘要 目的:探究上皮细胞膜蛋白2(EMP2)在甲状腺乳头状癌(PTC)中的表达及其对细胞侵袭迁移能力的影响。方法:收集我院2013年6月至2016年12月收治的120例行手术治疗的PTC患者肿瘤组织及相应癌旁组织,另收集16例PTC转移灶组织,荧光定量PCR(q PCR)及免疫组织化学(IHC)技术分别检测不同组织中EMP2 mRNA及蛋白表达情况。体外采用慢病毒转染针对EMP2的小干扰RNA载体,干扰人甲状腺乳头状癌细胞系TPC-1中EMP2的表达作为实验组细胞,转染空白对照载体作为对照组细胞,Western Blot检测两组细胞EMP2蛋白、上皮间充质转化(EMT)分子表达的差异,包括E-钙黏蛋白(E-cadherin)及波形蛋白(Vimentin),Transwell侵袭及迁移实验检测两组细胞侵袭迁移能力的差异。结果:EMP2 mRNA在PTC组织中表达显著高于癌旁组织,且转移灶组织表达显著高于非转移灶组织,差异存在统计学意义(P<0.05)。EMP2蛋白在PTC组织中表达阳性率显著高于癌旁组织,且转移灶组织阳性率显著高于非转移灶组织,差异存在统计学意义(P<0.05)。实验组细胞EMP2及Vimentin蛋白表达水平相比对照组细胞显著下调,差异存在统计学意义(P<0.05);而实验组细胞E-cadherin蛋白表达水平相比对照组细胞显著上调,差异存在统计学意义(P<0.05)。实验组穿透基质胶细胞数显著少于对照组细胞,差异存在统计学意义(P<0.01);实验组穿过小室孔的细胞数显著少于对照组细胞,差异存在统计学意义(P<0.01)。结论:EMP2在PTC组织中高表达,且在转移灶中相比非转移灶表达上调,干扰PTC细胞EMP2表达后其侵袭迁移能力显著下调,可能与抑制细胞EMT进程有关。 Objective: To investigate the expression of epithelial membrane protein-2(EMP2) in papillary thyroid carcinoma(PTC) and its effect on cell invasion and migration. Methods: The tumor tissue and corresponding para-cancerous tissues of 120 cases of PTC patients with surgical treatment in our hospital from June 2013 to December 2016 were collected,meantime 16 cases of PTC metastasis tissue were collected.Fluorescence quantitative PCR(qPCR) and immunohistochemistry(IHC) were used to detect the expression of EMP2 mRNA and protein in different tissues.In vitro,lentiviral transfection of small interfering RNA targeting vector of EMP2 was used to interfere the expression of EMP2 in human papillary thyroid carcinoma cell line TPC-1 as the experimental group cell,and blank control vector was transfected as the control group cell.Western Blot was used to detect the expression of EMP2 and epithelial mesenchymal transition(EMT) related molecule,including E-cadherin and Vimentin.Transwell invasion and migration experiment were used to detect the invasion and migration ability of two groups of cells. Results: The mRNA expression of EMP2 in PTC tissues was significantly higher than that in para-cancerous tissues,and the expression of metastasis tissues was significantly higher than that in non-metastasis tissues,and the difference was statistically significant( P 〈0.05).The positive rate of EMP2 protein in PTC tissues was significantly higher than that in para-cancerous tissues,and the positive rate of metastasis tissues was significantly higher than that in non-metastasis tissues,and the difference was statistically significant( P 〈0.05).The expression of EMP2 and Vimentin protein in experimental group cell were significantly lower than those in control group cell,while the expression of E-cadherin protein in experimental group cell was significantly higher than that in control group cell,and the difference was statistically significant( P 〈0.05).The number of experimental group cells through the matrigel was significantly less than the control group cells,the difference was statistically significant( P 〈0.01).The number of experimental group cells through holes was significantly less than the control group cells,the difference was statistically significant( P 〈0.01). Conclusion: EMP2 is highly expressed in PTC tissues and is up-regulated in metastatic tissues compared with non-metastatic tissues.After interfering the expression of EMP2 in PTC cells,the invasion and migration ability of PTC cells is down-regulated,which may be related to the inhibition of cell EMT progression.
