卒中后抑郁模型大鼠血清儿茶酚胺类递质变化的研究 被引量:3

摘要 目的建立卒中后抑郁(Post-stroke depression,PSD)大鼠模型,探讨卒中后抑郁大鼠血清中去甲肾上腺素(norepinephrine,NE)、肾上腺素(epinephrine,E)和多巴胺(dopamine,DA)含量的变化情况。方法选取大鼠48只纳入实验,随机分为4组,正常对照组12只,脑卒中组大鼠12只,孤养抑郁组大鼠12只,卒中后抑郁组12只。分别通过制备大脑中动脉栓塞模型,建立脑卒中组;给予慢性不可预测的温和刺激联合孤养法建立孤养抑郁模型。制备卒中后抑郁模型组时,首先建立大脑中动脉栓塞模型,成功制备卒中大鼠模型后,再通过慢性不可预测的温和刺激联合孤养法的综合造模方法进行二次造模,最后得到卒中后抑郁大鼠模型。应用高效液相色谱-荧光检测法检测各组大鼠血清中NE、E和DA的含量。结果 4组大鼠组间NE、E、DA含量比较,差异均有统计学意义(P<0.001)。两两比较发现,正常对照组、孤养抑郁组、脑卒中组、卒中后抑郁组的NE、E、DA含量有逐渐递减趋势,差异有统计学意义(P<0.05或0.01)。卒中后抑郁组大鼠血清中NE、E和DA含量变化均与大鼠体质量、糖水消耗量、水平运动得分呈正相关(P<0.001);而卒中后抑郁大鼠血清中NE、E和DA含量与大鼠直立运动得分、中央格停留时间、修饰次数相互关系不显著(P>0.05)。结论卒中后抑郁大鼠模型血清NE、E和DA含量明显下降,很可能与卒中后抑郁的发病机制有关。儿茶酚胺类神经递质水平的降低程度,与大鼠行为学改变程度高度相关。 Objective A rat model of post-stroke depression(PSD) was established to investigate the changes of serum norepinephrine( NE), epinephrine(E) and dopamine(DA) in rats with PSD. Methods 48 rats were selected and randomly divided into 4 groups : normal control group, stroke group, solitary depression group and PSD group,with 12 rats in each group. The model of middle cerebral arlery embolization was made to establish the stroke group, and the model of solitary depression group was established by chronic unpredictable mild stimulation combined with solitary raising method.In the preparation of the model of the PSD group, the middle cerebral arlery model was established at first.After successfully establishing the stroke rat model, and the model was established by chronic unpredictable mild stimulation combined with solitary raising method again. Finally,the rat model of the PSD group was obtained.And then,the contents of NE,E and DA in serum of rats in each group were detected by high performance liquid chromatography-fluorescence detection.Results Difference of the contents of NE,E and DA between the tour groups was statistically significant(P〈0.001).Pairwise comparison showed that the contents of NE,E and DA in the normal control group, the stroke group, the solitary depression group and the PSD group decreased gradually,and the difference was statistically significant(P〈0.05 or 0.01 ).In the PSD group, the changes of NE, E and DA contents were positively correlated with body mass, sugar water consumption and horizontal motor score in rats(P〈0.001 ), but the NE, E and DA contents were not significantly correlated with vertical movement scores, central lattice residence time and modification times (P〉0.05). Conclusion The levels of sermn NE, E and DA in the rats model of PSD significantly decrease ,which may be related to the pathogenesis of PSD.The decrease in the level of catecholamine neurotransmitters is highly correlated with the degree of ethological changes in rats.
