罗汉果内生菌中产环糊精葡萄糖基转移酶菌株的筛选及产酶条件优化 被引量:5

Screening of cyclodextrin glucosyltransferase-producing strains from endophytic bacteria in Siraitia grosvenorii and optimization of its enzyme-producing conditions

摘要 从罗汉果内生菌中筛选高产环糊精葡萄糖基转移酶(CGTase)的菌株进行菌种鉴定,并对其产酶条件进行优化。利用CGTase快速筛选法从罗汉果内生菌中筛选获得一株高产CGTase的菌株,编号为ND-6,经形态学和分子生物学鉴定其为一株解淀粉芽孢杆菌(Bacillus amyloliquefaciens)。通过单因素和正交试验优化得到其产CGTase条件为1.0%环糊精作碳源,1.0%蛋白胨作氮源,初始p H值为8.0,装液量70 m L/250 m L,发酵温度40℃。在此优化条件下,菌株所产CGTase酶活达到9 753 U/m L,是优化前的1.43倍。 The high cyclodextrin glucosyltransferase (CGTase)-producing strains from endophytic bacteria in Siraitia grosvenorii was screened and identified, and its enzyme-producing conditions were optimized. Using CGTase rapid screening method, a strain with the high enzyme activity was obtained and named ND-6, and the strain was identified as Bacillus amyloliquefaciens by morphology and molecular biology. Through single factor and orthogonal experiments, the optimum CGTase-producing conditions were using cyclodextrin 1.0% as carbon source and peptone 1.0% as nitro-gen source, initial pH 8.0, loading volume 70 ml/250 ml, and fermentation temperature 40 ℃. Under the optimal conditions, the CGTase activity of the strain could be up to 9 753 U/ml, which was 1.43 times higher than that of before optimization.
