摘要
本文以新疆伊吾县野山杏仁为原料,采用酶解法,以多肽水解度为考察指标,分析木瓜蛋白酶、中性蛋白酶及碱性蛋白酶以水浴酶解和磁力搅拌酶解方式,对野山杏仁多肽水解度的影响。采用全自动氨基酸分析仪测定杏仁多肽样本中水解氨基酸的种类和含量。选择1,1-二苯基-2-三硝基苯肼自由基(DPPH?)、羟基自由基(·OH)、超氧阴离子自由基(O^(2-)?),以VC为对照,测定不同浓度多肽液对这三种自由基的清除率,分析比较不同浓度野山杏仁多肽对三种自由基的清除能力,探讨野山杏仁多肽的体外抗氧化活性能力。实验结果表明:磁力搅拌酶解方式下利用木瓜蛋白酶多肽水解度为35.02%,为野山杏仁多肽水解最优方法;野山杏仁蛋白中富含丰富的氨基酸,是一类较为优质的植物蛋白资源;野山杏仁多肽具有清除自由基能力,其中对DPPH?自由基清除率最强为75.48%,有较强的抗氧化能力。
The effects of papain,neutral protease and alkaline protease,water bath enzymolysis and magnetic stirring on the degree of hydrolysis of wild almond peptide were analyzed by enzyme hydrolysis usingpeptide hydrolysis as the index. The raw materials of wild almond in Yiwu County of Xinjiang were studied by enzyme hydrolysis.The types and content of hydrolyzed amino acids in apricot polypeptide samples were determined by automatic amino acid analyzer. Compared with VC,1,1-diphenyl-2-picrylhydrazyl(DPPH),hydroxyl and superoxide anion were chosen to study wild almond polypeptides free radicals scavenging abilities. The experimental results showed that the hydrolysis degree by magnetic stirring papain enzyme was 35.02%,the best condition for wild almond peptide hydrolysis. Wild almond is rich in amino acids,and is a high-quality plant protein resources. Wild almond polypeptide has 75.48% DPPH scavenging ability and a high antioxidant ability.
