星点设计-响应面法优化丹参花粉离体萌发培养条件的研究 被引量:1

Study of in Vitro Cultural Condition for Pollen Germination of Salvia miltiorrhiza by Central Composite Design-Response Surface Method

摘要 以BK为基本培养基,在单因素试验的基础上,采用星点设计-响应面优化山东丹参、川丹参花粉离体培养,以花粉萌发率为响应指标,研究培养温度、蔗糖浓度、硼酸浓度与萌发率之间的响应面关系,优化花粉离体培养的条件。研究结果:温度、蔗糖浓度和硼酸浓度对山东丹参和川丹参花粉萌发率的影响均达到极显著水平(p<0.05),响应面优化后的最佳培养条件为:山东丹参BK+温度21.87℃+蔗糖151.20 g/L+硼酸273.83 mg/L,花粉萌发率的理论值为90.30%;川丹参BK+温度26.28℃+蔗糖133.81 g/L+硼酸237.71 mg/L,花粉萌发率的理论值为68.19%。验证得到二者萌发率分别为89.65%、66.31%。试验确立的数学模型的实测值与预测值相对偏差较小分别为-1.8%、-2.2%,可正确预测培养条件对花粉萌发率的影响,同时验证了星点设计-响应面优化丹参花粉离体培养条件优化方法模型的可靠性。该研究结果为今后进一步开展丹参花粉体外保存、人工控制授粉、选育优良杂交新品种以及川丹参不育机制的研究等提供了理论基础。 BK was used as the basic medium,and the in vitro culture of pollen from Salvia miltiorrhiza in Shandong and Sichuan was optimized by central composite design-response surface optimization method on the basis of single factor experiment.The pollen germination rate was set as the response index,and the response surface relationship among incubation temperature,sucrose concentration,boric acid concentration and germination rate was analyzed,so as to optimize the in vitro condition of pollen.The results showed that the effects of culture temperature,sucrose and boric acid concentration on the pollen germination rate were significant(p〈0.05).The optimal culture conditions after response surface optimization were as below:Salvia miltiorrhiza in Shandong:BK+temperature 21.87℃+sucrose 151.20 g/L+boric acid 273.83 mg/L,and the theoretical value for germination rate was 90.30% under this condition and the actual germination rate was 89.65%; Salvia miltiorrhiza in Sichuan:BK+temperature 26.28℃+sucrose 133.81 g/L+boric acid 237.71 mg/L,and the theoretical value for germination rate was 68.19% under this condition and the actual germination rate was 66.31%.It was obvious tha t the predicted value from an established mathematical model was close to the actual one,with relative deviation of-1.8% and-2.2%,respectively.This suggested that the model could be used to predict the effect of culture conditions on pollen germination rate.Meanwhile,it was proved that the model of central composite design-response surface optimization method in the optimization of in vitro culture condition for pollen germination of Salvia miltiorrhiza was reliable.This study could provide a theoretical basis for the further study on the preservation of pollen in vitro,the artificial control of pollination,the selection of new varieties of fine hybrids and the study of the mechanism of fertility of Salvia miltiorrhiza.
