摘要
目的探究叶酸对对氯苯丙氨酸(PCPA)抑郁模型大鼠的治疗作用及学习记忆能力的影响。方法将成年雄性SD大鼠30只随机分为对照、模型、模型+叶酸三组。通过腹腔注射PCPA建立抑郁模型,观察大鼠抑郁情绪,学习能力及下丘脑5-HT含量的变化。结果与对照组相比,模型组大鼠水迷宫2、3、4d训练平均逃避潜伏期[(19.44±4.04)s,(13.63±1.73)s,(11.13±2.78)s]明显高于对照组[(14.10±3.34)s,(5.60±1.04)s,(3.20±1.10)s](P<0.05);叶酸组大鼠水迷宫2、3、4d训练平均逃避潜伏期[(11.3±1.56)s,(8.06±1.62)s,(5.30±1.75)s],明显低于模型组[(19.44±4.04)s,(13.63±1.73)s,(11.13±2.78)s](P<0.05)。与对照组相比,模型组大鼠自主活动能力显著下降;悬尾和强迫游泳不动时间明显增长;联合皮质和海马脑区的5-HT含量明显减少,叶酸能够显著缓解以上变化。结论叶酸可显著改善模型大鼠的抑郁症状并提高模型大鼠的学习记忆能力,其机制可能与提高脑内5-HT含量有关。
Objective To access the effects of folic acid on learning and memory ability in PCPA depression rats. Methods Thirty SD rats were randomly divided into the control group, model group and folic acid group. The depression model was induced by intraperitoneal injection of PCPA. The changes in emotion, learning and memory behavior, and brain 5-HT were observed and compared among different groups. Results Compared with the model group, the immobility time of tail suspension test and force swimming test were obviously reduced. The spatial learning and memory ability was significantly decreased in Folic acid treatment group. folic acid treatment significantly increased 5-HT in the hippocampus and combined cortex. Conclusion Folic acid can significantly increase the emotion and learning ability in PCPA rats by increasing the 5-HT in hippocampus and combined cortex.
